Role of the Serotonin 5-HT2A Receptor in Learning

Abstract: This study reviews the role of the serotonin 5-HT 2A receptor in learning as measured by the acquisition of the rabbit's classically conditioning nictitating membrane response, a component of the eyeblink response. Agonists at the 5-HT 2A receptor including LSD (d-lysergic acid diethylamide) enhanced associative learning at doses that produce cognitive effects in humans. Some antagonists such as BOL (d-bromolysergic acid diethylamide), LY53,857, and ketanserin acted as neutral antagonists in that they had no e… Show more

“…Accordingly, we cannot exclude the possibility that SCH23390 has agonist actions at 5-HT2a and c receptors at the concentration used (see Porter et al 1999). However, the activation of these receptors has been implicated in the facilitation of hippocampusdependent memory (Buhot et al 2000;Harvey 2003), in contrast to the findings reported here.…”

“…Similarly, a recent MMN study with S-ketamine and the 5-HT 2A R agonist DMT showed that the frontal MMN was only affected by S-ketamine and not by DMT (Heekeren et al, 2008). Although 5-HT 2A R seems to be involved in different forms of working memory (Williams et al, 2002) and 5-HT 2A R agonists such as LSD or DOM enhanced associative learning as measured by eyeblink conditioning in rabbits (Harvey, 2003), the implicit perceptual learning as indexed by the frontal memory trace effect appeared not to be sensitive to manipulations of the 5-HT 2A R system (or to the dose of psilocybin used in this study). Although it has been proposed that neuromodulators like serotonin may influence the processing of PE (Corlett et al, 2009) and may be one of the key modulators interacting with NMDARs to produce aberrant synaptic plasticity in schizophrenia ), this does not necessarily contradict our findings since such a putative role of serotonin could be expressed in a regionally specific manner (ie, in other circuits than the auditory system), and may thus not be a critical factor in PE processing during the MMN paradigm (cf.…”

“…Increases in 5-HT2A receptors reinforce learning and memory; conversely, receptor antagonists have a negative impact on learning (56). The 5-HT2A receptor antagonist volinanserin MDL 100907 (MDL) increases sleep and decreases wakefulness in rats.…”
