Role of lipopolysaccharide in assembly of Escherichia coli outer membrane proteins OmpA, OmpC, and OmpF

Abstract: Selection was performed for resistance to a phage, Ox2, specific for the Escherichia coli outer membrane protein OmpA, under conditions which excluded recovery of ompA mutants. All mutants analyzed produced normal quantities of OmpA, which was also normally assembled in the outer membrane. They had become essentially resistant to OmpC and OmpF-specific phages and synthesized these outer membrane porins at much reduced rates. The inhibition of synthesis acted at the level of translation. This was due to the pre… Show more

“…Consistent with tolerance to cations, ompC and ompA, which encode outer-membrane porins (Ried et al, 1990), are reduced in most of the adapted strains. b0795, which shows similarity to CzcB (a component of a cation efflux pump ; Nies et al, 1989), is reduced in all four strains.…”

“…Modifications in LPS structure have been associated with changes in assembly and composition of OM proteins (Ames et al, 1974;Ried et al, 1990;de Cock et al, 1999), in A. brasilense Sp7 also (Lerner et al, 2009). Therefore, we examined whether the mutant strains differed in their OM protein profiles relative to the wild-type.…”

“…Several reports have documented the effect of LPS on the biosynthesis, folding and function of porins. E. coli mutants expressing defective LPS molecules of the 'deep rough' type have very low amounts of OmpF and OmpC porin in their outer membrane [11]. Sen and Nikaido examined E. coli spheroplasts secreting water-soluble, monomeric OmpF.…”
