Role of Interleukin-6 in Depressive Disorder

Abstract: Major depressive disorder (MDD), which is a leading psychiatric illness across the world, severely affects quality of life and causes an increased incidence of suicide. Evidence from animal as well as clinical studies have indicated that increased peripheral or central cytokine interleukin-6 (IL-6) levels play an important role in stress reaction and depressive disorder, especially physical disorders comorbid with depression. Increased release of IL-6 in MDD has been found to be a factor associated with MDD pr… Show more

“…In a study, IL-6 knockout mice became resistant to the development of depression-like symptoms (Monje et al, 2011 ). The underlying mechanisms involve in two pathways, the HPA axis and neurotransmitter metabolism, both of which are affected by increased IL-6 in depression (Ting et al, 2020 ). Furthermore, anxious patients also had higher serum levels of IL-6 than common people (Tang et al, 2018 ; Zou et al, 2020 ).…”

“…It was apparent that there is an inverse correlation between the serum IL-6 level and prefrontal cortex thickness. Sleep disturbance and impairment in sustained attention was positively related to an increase in IL-6 level ( 34 ). Patients with cancers such as lung, ovarian, colorectal, pancreatic, and others show higher levels of IL-6 compared to healthy controls.…”

“…It was found that improvement in depressive symptoms in these patients was correlated with normalization of IL-6 levels. Symptoms such as anorexia, insomnia, and fatigue were significantly associated with elevated IL-6 levels ( 34 ).…”

“…As two of the most common types of psychological disorders, depression and anxiety had caused millions of morbidity and mortality each year all over the world [6]. It was confirmed that many biological [7,8], genetical [9], psychological [10], and social environmental [11] factors are involved in the pathogenesis of depression and anxiety. Psychological disorders can often affect one's physical health in a variety of known or unknown ways.…”
