Nepeta pogonosperma is an important medicinal plant with anti-inflammatory effects. An efficient and reliable transformation system for this plant was developed through optimization of several factors which affected the rate of Agrobacterium rhizogenes mediated transformation. Five bacterial strains, A4, ATCC15834, LBA9402, MSU440 and A13, two explant types, leaves and stems, and several co-cultivation media were examined. The maximum rate of hairy root induction was obtained from stem explants using MSU440 and ATCC15834 bacterial strains. A drastic increase in the frequency of transformation (91 %) was observed when MS medium lacking NH 4 NO 3 , KH 2 PO 4 , KNO 3 and CaCl 2 . Hairy root lines were confirmed by polymerase chain reaction (PCR) using primers of the rolB gene. According to Southern blot analysis, one T-DNA copy was inserted into each of the hairy root lines. In the present study, transgenic hairy roots have been obtained trough genetic transformation by A. rhizogenes harbouring two plasmids, the Ri plasmid and pBI121 binary vector harbouring gus reporter gene. Expression of the gus gene in transgenic hairy root was confirmed by histochemical GUS assay.

The study was conducted to determine the phytochemical constituents and also to assess the toxicity of the aqueous extract of Boswellia dalzielli stem bark on the liver and kidney of male mice. Healthy male mice weighing between 45 to 50 g were used. The mice were grouped into four different groups, each containing four mice, respectively. Different concentrations (200, 400 and 600 mg/kg) of the aqueous extract of B. dalzielli stem bark were orally administered, respectively according to group, at the interval of 12 h per day, whereas the other group was not given any treatment and this served as the normal control, providing a baseline data. The experiment lasted for two weeks after which the animals were sacrificed by human decapitation. The serum was obtained and used for the analysis of the parameters of hepatic and renal functions, respectively. The results of the phytochemical screening revealed the presence of alkaloid, flavonoid, tannins, saponins, balsam, cardiac glycoside, terpenes, steroids, resins, and phenols. All the treatment groups showed an elevated serum aspartate transaminase (AST) and a low serum alanine transaminase (ALT), and ALP was (p < 0.05), indicating possibly a damage to the liver. Also, the treatments showed a low serum urea and a normal creatinine level (p < 0.05). Therefore, it was concluded that the extract had some toxic effects on the liver but had no toxic effect on the kidney of the mice.

An Efficient Method for Direct Shoot Regeneration from Leaf Explants of Solanum nigrum Induced by Thidiazuron

Direct shoot regeneration was established from leaf, petiole and nodal explants from in vitro and field grown plants of Solanum nigrum on Murashige and Skoog (MS), and Gamborg's (B5) media containing different levels of plant growth regulator, Thidiazuron (TDZ). Comparison of the individual effect of basal media, levels of TDZ, explant type or source of explants on shoot regeneration was tested. Percent shoot regeneration from leaf explant was highest (86.66%) followed by in vitro source of explant (83.65%), B5 basal medium (83.20%) or 2.27 µM TDZ (73.70%) after 60 days of initial culture. The number of shoots was highest (35.10) with 9.13 cm longest shoot at 2.27 µM TDZ. There was a little correlation between predictors and dependent variables was found; whereas, GLM analysis demonstrated significant correlation between the factors. In vitro shoots were transferred to a half-strength MS medium containing different concentrations of NAA or IBA in MS or B5 medium. MS medium produced 100% roots supplemented with 0.5 µM NAA after 28 days. However, stout and hardy rooted shoots were obtained at 0.5 µ IBA with 10.19 mean number and 5.57 cm long roots. Rooted shoots were shifted to ex vitro conditions in polyethylene-cups filled with peat moss: sand: garden soil (1:1:1) with 85.50% survival with true-totype morphology under the natural conditions. The present investigation demonstrated that TDZ may be used for high frequency direct shoot regeneration from leaf explants of S. nigrum on B5 basal medium. This protocol may be useful for subsequent high yielding metabolites producing plants.
