Streptomyces sampsonii KK1024 having strong chitinolytic activity was isolated from crab-shell rich soil at Muan, Jeolanamdo. The KK1024 produced chitinase, protease, gelatinase and lipase. When 50% of KK1024 culture broth was treated to juveniles and eggs of root-knot nematode, juvenile mortality at 3 days was 81.67% and egg hatch rate at 5 days was 2.00%. When 183.7 μg mL -1 of crude enzyme produced by KK1024 was treated, juvenile mortality at 3 days was 96.00% and egg hatch rate at 5 days was 5.33%. At 1% of butanol extract from KK1024, juvenile mortality was highest with 90.00% and egg hatch rate was lowest with 0%. The comparison of the effect of KK1024 culture broth with only medium, synthetic fertilizer, and commercial nematicide on tomato growth and nematode infection was examined in pot trials. KK1024 culture broth showed lower number of egg mass and gall in plant, and population of juveniles in soil compared with only medium and synthetic fertilizer treatment, but not in commercial nematicide. However, the highest shoot weight and length was discovered in KK1024 culture broth. These results suggest that Streptomyces sampsonii KK1024 producing lytic enzymes and nematicidal compounds can be one of candidates for biocontrol agents against root-knot nematodes.

Abstract-Several Streptomyces species are known to produce metabolites that inhibit plant pathogens. One such compound is geldanamycin (GA), a benzoquinone ansamycin originally isolated from Streptomyces hygroscopicus. We examined the effect of geldanamycin on egg hatch and juvenile motility in Caenorhabditis elegans and in two populations of the plant-parasitic nematode Heterodera glycines. When C. elegans eggs were exposed to geldanamycin, both hatch and motility were reduced by GA doses between 2 and 50 mg/ml. The H. glycines inbred populations TN17 and TN18 exhibited low dose stimulation of hatch and motility, whereas levels occurring at higher GA doses were at or below control levels. These experiments represent the first demonstration of geldanamycin effects in C. elegans and H. glycines and suggest that the heat shock chaperone Hsp90, the known molecular target of geldanamycin, may be involved in nematode egg hatch and motility. This study also indicates that geldanamycin-producing strains of Streptomyces may be useful as biocontrol agents for nematodes.

Growth promotion and disease control by rhizobacteria are complex interrelated processes that involve direct and indirect mechanisms. The mechanisms include synthesis of some metabolites (auxin, cytokinin and gibberellins), induction of 1-aminocyclopropane-1-carboxylate (ACC) deaminase, production of siderophore, antibiotics, hydrogen cyanide hydrogen cyanide (HCN) and volatile compounds. They also include mineral solubilization competition, and induction of systemic resistance. These bacteria are suitable as soil inoculants because they have the potential for rapid and aggressive colonization. This feature alone is characterised as a disease control mechanism, which prevents the invasion of detrimental soil microorganisms onto the root surface. Inoculant-based plant growthpromoting rhizobacteria (PGPR) is applied extensively on agricultural crops to improve plants' growth and simultaneously reduce chemical inputs like fertilizer and pesticide which can cause environmental degradation. The structure of the rhizobacterial community is affected by several factors including plant genotype and is determined by the amount and composition of root exudates. In addition, soil type and fertility are the contributing factors that shape the community. This form of communication can affect plants' growth, nutrient status and also susceptibility to stress and pathogens in the host plant. PGPR inoculants cause diverse beneficial interactions among plants, which leads to sustainable and environment-friendly agriculture. The application of rhizosphere soil of agricultural crops with desirable bacterial populations is considered promising in both laboratory and greenhouse experiment. Further, a clearer understanding of the way PGPRs promote plants' growth can lead to expanded exploitation of these 'biofertilizers' in order to reduce the potential negative environmental effects associated with food and fiber production.
