Role of autologous mesenchymal stem cells associated with platelet-rich plasma on healing of cutaneous wounds in diabetic mice

Abstract: Both treatments gave better results than daily cleaning of the cutaneous lesions with saline or covering of the lesions with semipermeable adherent bandage.

“…The cells were isolated from the bone marrow of healthy, expanded in vitro rabbits submitted to osteogenic and chondrogenic cell differentiation assay and aliquoted at the concentration of 1x10 6 cells/mL for immediate use, according to methodologies previously described by Argolo Neto et al 9,16 and Carvalho et al 10 .…”

“…Three aliquots of 1x10 4 MSC were maintained in complete F12 DMEM medium, together with a 3 mm sample of sterile hemostatic sponge of hydrolyzed collagen, in a six-well culture plate, incubated at 37°C, 5% CO 2 and 95% moisture 16 .…”

“…CG rats showed complete healing with a mean time of 14.75 days, DG rats had complete healing with a mean of 21.25 days and, in MSCDG rats, the healing process was completed on a mean time of 13.5 days. In view of the clinical evidence of improvement in the wound healing in animals treated with MSC, it is believed that the benefits for the quality of healing resulting from the use of mesenchymal stem cells may be better confirmed at evaluations carried out after 15 days, when a better cosmetic scar quality could be assessed by the organization of collagen deposition (ARGOLO-NETO et al, 2012;CASADO et al, 2014). In this study, the MSC therapy was effective in accelerating the healing process and increasing the formation of collagen fibers, similar to observed by CASADO et al (2014) andCHUNMENG et al (2004).…”

“…Six rats from each group (DG and MSCDG) were chosen at random to have the wounds photographed daily throughout the observation period for the evaluation of wound area (WA) and healing time (HT), according to the methodology of ARGOLO-NETO et al (2012). Images were transferred to a computer and wound area was measured using ImagePro Plus ® image analysis software.…”
