Role of 5‐HT1A autoreceptors in the mechanism of action of serotoninergic antidepressant drugs: recent findings from in vivo microdialysis studies

Abstract: Although a new generation of selective serotonin reuptake inhibitors (SSRIs) has been introduced in therapeutics as antidepressant drugs, a two to four week lag period still occurs between starting treatment with SSRIs and the onset of therapeutic effects in man. In vivo cerebral microdialysis can be used to measure extracellular concentrations of serotonin (5-hydroxytryptamine, 5-HT), which reflect intrasynaptic events. With the coupling of this new experimental method to very sensitive analytical assays such… Show more

“…5 Neuroplastic effects that include downregulation of 5-HT 1A autoreceptors and increased synaptic 5-HT content are involved in the onset of antidepressant efficacy. 6,7 These effects are only seen with chronic but not with acute fluoxetine administration. 6,8,9 In this regard, chronic fluoxetine administration upregulates 5-HT 2C receptors in the rat choroid plexus, 10 increases rat brain 5-HT 2A receptor signaling 11 and increases 2,5-dimethoxy-4-iodoamphetamine-induced cFOSÀ gene expression.…”

“…The raphe nuclei are a group of neuronal clusters responsible for serotoninergic signalling within the brain and are involved in mood (Gardier et al, 1996) and circadian rhythms (Monti, 2010). The perirhinal cortex maintains multiple connections throughout the raphe nuclei (Vertes, 1991;Hermann et al, 1997;Vertes et al, 1999).…”

“…On chronic administration, SSRIs have been shown to desensitize 5-HT 1A autoreceptors (De Montigny et al, 1990;Le Poul et al, 1995;Davidson and Stamford, 1998). This desensitization, which allows levels of synaptic 5-HT in the forebrain to rise (Gardier et al, 1996;Dawson et al, 2000), shows some temporal correspondence with the onset of antidepressant efficacy (Gardier et al, 1996;Le Poul et al, 1995) and hence is accepted to be an important feature of the response to chronic administration of SSRIs (see Blier et al, 1987, Blier, 2001Artigas et al, 2001).…”
