Recently, additional long noncoding RNAs (lncRNAs) have been identified and their possible roles were investigated in a variety of human tumors. One of these lncRNAs, LINC01929, promoted the progression of some cancers, whereas its expression and biological function in human oral squamous cell carcinoma (OSCC) remains still mostly uncertain. The LINC01929 expression in OSCC tissues or cell lines was identified via quantitative real-time polymerase chain reaction. The cell counting kit-8, transwell migration, wound-healing, and flow cytometry assays were utilized to characterize the functions of LINC01929 in OSCC cells. The interactive relationships between LINC01929 and miR-137-3p, miR-137-3p and Forkhead box C1 (FOXC1) were investigated by the dual-luciferase activity assay. Our findings demonstrated that LINC01929 was highly expressed in OSCC tissue samples and cell lines, whereas miR-137-3p expression was downregulated. LINC01929 acted as a carcinogenic lncRNA with accelerated OSCC cell proliferation, migration and invasion, and suppression of apoptosis. We further indicated that LINC01929 facilitated tumor growth in xenograft mouse models. Mechanistically, LINC01929 acted as a sponge for miR-137-3p to elevate FOXC1 expression, which is the target of miR-137-3p. In addition, downregulated miR-137-3p expression rescued the suppressive behaviors of LINC01929 knockdown on the biological behaviors of OSCC cells. Taken together, LINC01929 functioned as a tumor-promoting lncRNA via the miR-137-3p/FOXC1 axis in OSCC, suggesting novel targets for OSCC therapy.

Background: Osteosarcoma is one of the most common malignant neoplasm among children and adolescents. Studies have shown that metabolism-related pathways are more important for the development and metastasis of osteosarcoma. Long non-coding RNA (LncRNA) plays a key role in the occurrence and progression of cancer in a variety of ways, Metabolism-related lncRNA-mediated molecular mechanisms in the regulation of osteosarcoma have not been fully elucidated.Methods: In this study, all metabolic-related mRNAs and metabolic-related LncRNA in osteosarcoma were extracted and identified based on transcriptomic data from the TCGA database. The survival analysis, The univariable and multivariable independent prognostic analysis, The results of gene set enrichment analysis (GSEA) and the nomogram were used to construct a prognosis signature with metabolic LncRNA as prognostic factor.Results: 9 prognostic factors including that LncRNA AC009779.2, LncRNA AL591895.1, LncRNA AC026271.3, LncRNA LPP-AS2, LncRNA LINC01857, LncRNA AP005264.1, LncRNA LINC02454, LncRNA AL133338.1 and LncRNA AC135178.5, respectively. The survival analysis showed that the difference in expression of an individual LncRNA was closely related to poor prognosis in osteosarcoma. The univariable and multivariable independent prognostic analysis showed that the signature had good independent predictive ability for patient survival. The results of gene set enrichment analysis (GSEA) suggest that these predictors may be involved in the metabolism of certain substances or energy in cancer. The nomogram is further drawn for clinical guidance and assistance in clinical decision-making.Conclusions: This study identified multiple metabolic-related lncRNA that can be considered as novel therapeutic targets for osteosarcoma and contribute to better exploring the specific regulatory mechanisms of lncRNA in the metabolism of osteosarcoma.

MicroRNAs (miRNAs or miRs) are non-coding small RNAs that target specific messenger RNAs to inhibit protein translation. miR-200a and miR-141 function as tumor suppressors by targeting STAT4. These two miRNAs belong to the same family, and their expression is often decreased in various cancer types, but are located on different chromosomes of the human genome. The present study showed that the expression levels of miR-141 and miR-200a in serum and cells of liver cancer are significantly downregulated. The expression levels of miR-141 and miR-200a are closely associated with clinicopathological features of liver cancer, especially metastasis and invasion. It is first reported that STAT4 is the new common target gene of miR-141 and miR-200a. In the present study, miR-141 and miR-200a were confirmed to inhibit the expression of E-cadherin and vimentin synergistically during epithelial-mesenchymal transition to regulate the proliferation, migration and invasion of liver cancer cells by targeting STAT4. Simultaneous overexpression of miR-200a and miR-141 resulted in stronger effects compared with each miRNA alone. In addition, overexpression of STAT4 significantly reversed the tumor suppressive roles of miR-200a and miR-141 in liver cancer cells. These findings enrich the tumor suppressor mechanisms of the miR-200 family, and may also provide new experimental and theoretical basis for the use of miRNAs for early diagnosis, prognosis and thorough treatment of liver cancer.
