Risk of Alzheimer's Disease Biological Misdiagnosis Linked to Cerebrospinal Collection Tubes

Abstract: Tau proteins and amyloid-β (Aβ) peptides are the current recognized cerebrospinal fluid (CSF) biomarkers used as an aid in the diagnosis of Alzheimer's disease (AD). However, there is no consensus on their clinical use due to non-qualified cut-off values, probably related to the observed high pre-analytical and analytical variability. Standardized pre-analytical protocols have therefore been proposed. Importantly, these recommend the use of polypropylene collection/sampling tubes while, to date, no broad compa… Show more

“…The fact that long-term studies show stable concentrations for all 3 biomarkers means that differences between liquid N 2 , Ϫ80°C, and Ϫ20°C are most likely attributable to the initial freezing conditions. This is also seen for tubes that immediately adsorb A␤ (25 ).…”

“…Very impressive are the results of a study published last year that dealt with the influence of the material of the sample tubes on the measurement results [16,17] . The concentrations of tau protein, phospho-tau protein and A β 1 -42 were investigated in dependence on the sample tubes used, for a total of 11 different types of tubes.…”

“…The consequences on AD biological diagnosis could be very important, as demonstrated in a multisite study conducted in four French Centers for Memory Research and Resources (CMRR) (Montpellier, Lille, Lyon and Paris) [33]. Standardization of the collection tubes (using, e.g., the tube Sarstedt 62.610.201) could improve multicenter comparison and decrease the risk of wrong diagnosis [34].…”
