Ribonucleic Acid-Dependent Deoxyribonucleic Acid Polymerase in Visna Virus

Abstract: A ribonucleic acid-dependent deoxyribonucleic acid polymerase was found in virions of visna virus. The enzyme product was resistant to ribonuclease and alkaline hydrolysis but susceptible to the digestion of deoxyribonuclease. Visna virus is a medium-sized, ether-sensitive virus of sheep (8). It is formed by budding of the cytoplasmic membrane of the host cells and contains ribonucleic acid (RNA) as demonstrated by radioactive labeling of purified virus (2; Lin and Thormar,Virology, in press), but its multipli… Show more

“…It contains a poly(A) segment of large and homogeneous size (9). The virus particle, whose structure is very close to that of RNA tumor viruses (18,26), contains an RNA-dependent DNA polymerase (15,20,23) and replicates through the intermediate of a DNA provirus (11). Lastly, Visna virus has been reported to transform murine cells in vitro (25).…”

“…VOL. 15,1975 After purification, an aliquot of the virus suspension was centrifuged in a 10 to 50% linear potassium tartrate gradient (data not shown). The virus banded at a density of 1.17 g/ml, a density identical to that of older virus.…”

“…RNA viruses containing an RNA-directed DNA polymerase (RDDP) have been associated with a variety of chronic infections in animals, including neoplasms (26) and encephalitis (27,28). Of particular interest are the reports that oncogenic RNA viruses can be implicated in the immune complex glomerulonephritis of New Zealand black mice (29) and possibly in systemic lupus erythematosus (2,30).…”

“…Many lytic and tumorigenic viruses contain nucleic acid-polymerizing enzymes in their virions (3,4,11,16,24,26,28). Little information is available concerning regulatory mechanisms of virion-associated enzyme activities during virus replication and tumorigenesis.…”
