Rhynchophylline ameliorates myocardial ischemia/reperfusion injury through the modulation of mitochondrial mechanisms to mediate myocardial apoptosis

Abstract: Rhynchophylline (RP), the primary active ingredient of Uncaria rhynchophylla, has an anti-hypertensive effect and protects against ischemia-induced neuronal damage. The present study aimed to examine the roles and mechanisms of RP in myocardial ischemia-reperfusion (MI/R) injury of rat cardiomyocytes. Cell viability, reactive oxygen species, mitochondrial membrane potential (MMP) and cell apoptosis were examined by a Cell Counting Kit-8 assay and flow cytometry, respectively. An ELISA was performed to assess t… Show more

“…Uncaria rhynchophylla , a traditional Chinese medicine, is usually used in traditional Chinese medicine formulas for the treatment of asthma. Rhynchophylline (Rhy), a kind of indole alkaloid isolated from Uncaria rhynchophylla , shows anti-inflammation and anti-oxidant capabilities [ 6 , 7 ]. Our previous study showed that rhynchophylline can mitigate inflammation in asthma, attenuate hypertrophy of bronchial smooth muscle cells [ 8 ], however, the effect of rhynchophylline on autophagy in asthma remains unknown.…”

“…In similar experimental conditions, U. Rhyncophylla has been shown to favor anti-apoptotic over pro-apoptotic proteins in vitro [ 207 ]. Moreover, Rhy, Isorhy and U. Rhyncophylla were all shown to prevent the increase of caspase-3 cleavage in various models of neurotoxicity [ 70 , 79 , 94 , 98 , 207 , 208 ]. The cleavage of caspase-3, known as an ‘executor of apoptosis’, is often considered the ultimate step in the apoptotic cascade [ 209 ].…”
