Rhinovirus-Induced MMP-9 Expression Is Dependent on Fra-1, Which Is Modulated by Formoterol and Dexamethasone

Abstract: Matrix metalloproteinase-9 is implicated in airway inflammation and airway remodeling in asthma. We have previously confirmed that human rhinovirus-16 (HRV-16) infection increases MMP-9 expression both in vivo and in vitro. However, the role of the AP-1 sites within the MMP-9 promoter and the effect of commonly used asthma pharmacotherapies in modulating human rhinovirus (HRV)-induced MMP-9 production have not yet been elucidated. Experiments were performed in vitro in the human bronchial epithelial (HBE) cell… Show more

“…In vitro, we and others have found that steroids inhibit rhinovirus [47][48][49] and RSV-induced cytokine release, supporting the notion that the use of steroids in vivo would suppress virus-induced inflammation. Interestingly, data from in vitro studies have suggested that the timing of administration of steroids has profound effects upon their ability to inhibit virus-induced cytokines.…”

“…This result indicates that another signaling pathway regulates ERK-mediated IL-6 expression. ERK is involved in many signaling pathways activated during immune responses, such as those associated with IL-8 expression (Pera et al, 2012), and asthma (Tacon et al, 2012;Yadav et al, 2006). Based on that ERK result, we modified our hypothesis.…”

“…Cyclic AMP/PKA-dependent, gene-targeted, anti-inflammatory mechanisms are also operative at several levels, including: i) antagonism of transcription factors;51,52 ii) induction of synthesis of the anti-inflammatory cytokine, interleukin (IL)-10;3,5357 and iii) potentiation of glucocorticoid (GC)/glucocorticoid receptor (GR)-activated gene transcription, which contributes to the beneficial therapeutic interactions of LABAs with ICS 5874…”
