RGD-Conjugated Dendrimer-Modified Gold Nanorods for in Vivo Tumor Targeting and Photothermal Therapy

Abstract: Successful development of safe and effective nanoprobes for tumor targeting and selective therapy is a challenging task. Although gold nanorods(GNRs) have the potential to perform such a role, the toxicity of surfactant cetyltrimethylammonium bromides (CTAB) on their surfaces limits their applications. Here, polyamidoamine dendrimer was applied to replace CTAB molecules on the surface of gold nanorods. When the resultant dendrimer-modified gold nanorods conjugated with arginine-glycine-aspartic acid (RGD) pept… Show more

“…RGD peptide can mediate cell adhesion and proliferation and plays an important role in tumor angiogenesis and metastasis, being considerably upregulated in the endothelium during angiogenesis. Therefore, RGD represents a marker for malignancy [18] and has become a useful tool for the targeting of drugs and probes for functionalized imaging contrast agents [19,20].…”

“…It was reported that the systemic TNF toxicity of Aurimune was decreased dramatically compared with TNF alone (For example, the fever side effect was managed) and the target effect was realized. Another product, AuriTolℱ (CYT-21001), also from Cytlmmune Sciences, has reached the preclinical stage [46][47][48][49][50].…”

“…They found that GNRs showed over 20% accumulation in tumor tissues and the tumors were completely resorpted within 6 days after two times of laser irradiation. Li et al [65] also investigated RGD peptide conjugated GNRs for in vivo photothermal therapy. The results showed that the tumor completely disappeared after four times irradiation within one month.…”
