RGD and BMP-2 mimetic peptide crosstalk enhances osteogenic commitment of human bone marrow stem cells

Abstract: For a long time, scientists thought that the differentiation of Mesenchymal Stem Cells (MSCs) into bone cells was dictated by growth factors. This manuscript shed light on other ligands that play a crucial role in regulating MSCs fate. In concrete terms, it was demonstrated that the osteoinductive effect of BMP-2 peptide is 2 folds improved in the presence of adhesive RGD peptide. Compared to previous works highlighting this synergistic cooperation between RGD and BMP-2 peptides, the main strength of this work… Show more

“…The spatial patterning of peptides was ascertained at different stages, before and after peptide grafting and patterning. In a previous study, the covalent grafting of fluoro‐tagged RGD‐TAMRA and BMP‐2‐FITC peptides was evidenced using X‐ray photoelectron spectroscopy (XPS) . The peptides were randomly immobilized onto glass substrates.…”

“…Given that the surface roughness has been recognized for long time to greatly affect stem cells behavior and functions, we previously measured the surface roughness of RGD‐TAMRA and BMP‐2‐FITC peptides distributed in homogenous way on glass surface . Results showed no significant difference in the surface roughness between these two conditions.…”

“…Advancements in bone tissue engineering require the improvement of tissue scaffolds that not only have large surface areas and highly inter‐connected porous structures with suitable porosity and pore size to support cell attachment and proliferation, but also carry signaling molecules that mediate cell activity . Biodegradable polymer scaffolds, such as chitosan (CS) and poly( l ‐lactide‐ co ‐glycolide) scaffolds, have been widely used because of their excellent biocompatibility and low toxicity.…”

“…Growth factor derived peptides at high density have been shown to accommodate receptor activation similar to full-length proteins, while when presented at low density they may inhibit growth factor signaling. 2730 These studies underscore the importance of understanding the relationships associated with ligand presentation and receptor signaling, when working with surface presenting short peptides.…”
