RFLP analysis of SLA haplotypes in Swiss large white and American Hampshire pigs using SLA class I and class II probes

Abstract: Restriction fragment length polymorphism (RFLP) analyses of swine leukocyte antigen (SLA) class I and class II genes from Swiss Large White and American Hampshire families were performed using porcine DNA probes. Class I and class II RFLPs associated with the serologically-defined haplotypes SLA H1, H8, H16 and H24 and with serotypes SLA 15, 16; SLA 14; and SLA 6, SB 19, were identified. Seven allelic class I RFLP patterns were observed. For genes in the SLA class II region, six allelic RFLP patterns of DQA an… Show more

“…Low correlation between serogical haplotypes and RFLP fragments has been reported for man (e.g. Driesel et al 1985), cattle (Lindberg & Andersson 1988;Oliver et al 1989) and was also indicated by earlier studies in pig (Chardon et al 1985;Shia et al 1991). It is obvious that these two methods detect polymorphisms in different regions of the class I genes or even in different genes of the class I gene region.…”

“…-). Furthermore, RFLP analysis with EcoRI of haplotypes H04, H01, H16 and H24 using long coding region probes (Chardon et al 1985;Shia et al 1991) detected 7, 8/9, 10 and 9/10 fragments, respectively.…”

“…The pdDRB DNA sequences share 78-82% similarity to human leucocyte antigen (HLA)-DRB, and pdDQB DNA sequence shows 80% (exon 2) and 87% (exon 3) identity to HLA DQB sequences. Southern blotrestriction fragment length polymorphism (RFLP) analyses showed DQB and DRB to be polymorphic in NIH miniature pigs (Sachs et al 1988) and in the Swiss Large White and American Hampshire breeds (Shia et al 1991). Five alleles for exon 2 Shia, Bradshaw, Rutherford, Le win, Schook The polymerase chain reaction (PCR)-RFLP typing method has been used to detect polymorphism at the second exon (PI) of the HLA-DQA1, -DQBl, -DRBZ, -DRB3, -DRB4 and -DRB5 loci (Uryu ef al.…”

“…Among the peculiarities reported years ago are the remarkably small number of detectable MHC class I sequences. Depending on the techniques used, their extent varied from seven to ten per haplotype in miniature lines as well as in commercial breeds (8)(9)(10)(11). In comparison with pigs, humans and rodent species show several tens of class I sequences.…”
