RF/6A Chorioretinal Cells Do Not Display Key Endothelial Phenotypes

Abstract: PurposeThe misuse of inauthentic cell lines is widely recognized as a major threat to the integrity of biomedical science. Whereas the majority of efforts to address this have focused on DNA profiling, we sought to anatomically, transcriptionally, and functionally authenticate the RF/6A chorioretinal cell line, which is widely used as an endothelial cell line to model retinal and choroidal angiogenesis.MethodsMultiple vials of RF/6A cells obtained from different commercial distributors were studied to validate… Show more

“…The monkey choroidal EC line RF6/A and human retinal microvascular endothelial cells (HRMECs) are widely used as choroidal and retinal EC models, respectively. Since a recent study showed that the RF6/A cells do not exhibit key EC features 17 , we used HRMECs to investigate the role of AIBP in cholesterol efflux in retinal endothelial cells. HRMECs were incubated with control media, AIBP, HDL 3 (a subfraction of HDL, which is an efficient cholesterol acceptor) 13 , or AIBP in combination with HDL 3 .…”

“…Human retinal microvascular endothelial cells (HRMECs) were purchased from Cell Systems Corporations (catalog no. ACBRI 181; Kirkland, WA, USA) 19,20 and cultured in M199 medium supplemented with 20% fetal bovine serum, 3 ng/mL FGFbasic, 10 units/mL heparin, and 1% streptomycin/penicillin at 378C under 5% CO 2 , 95% air. D-glucose was added to the medium at a final concentration of 30 mmol/L to generate highglucose (HG) conditions.…”

“…RF/6A cells were used for the tube formation assay as described previously in detail. 24 Since an interesting study reported that RF/6A lost some key EC markers, 25 we compared its angiogenic reactions to VEGFA with human microvascular ECs (HMVECs) and got identical responsiveness. Finally, as RF/ 6A is the most prevalent in vitro choroidal and retinal EC model, 26,27 we decided to include it in our tube formation experiment.…”
