Retention mechanism of pH‐peak‐focusing in countercurrent chromatographic separation of baicalin and wogonoside from Scutellaria baicalensis Georgi

Abstract: An efficient and target-oriented pH-peak-focusing countercurrent chromatographic method was established for large-scale separation of baicalin and wogonoside from the crude exact of traditional Chinese medicinal herb Scutellaria baicalensis Georgi. An optimized two-phase solvent system composed of n-butanol-ethyl acetate-methanol-water (1:4:0.5:5, v/v) was selected. Trifluoroacetic acid (10 mmol/L) was added to the upper organic phase, used as the stationary phase. One liter of the aqueous lower phase was used… Show more

“…Furthermore, the excessive use of organic solvents is harmful to the environment. However, as a liquid–liquid partition chromatography, high-speed countercurrent chromatography (HSCCC) may use a liquid stationary phase without a solid support, thereby avoiding irreversible adsorption and benefiting complete sample recovery [ 59 , 60 ]. HSCCC has been successfully used to separate the antitumor compound antroquinonol from the solid-state cultured mycelia of A .…”

“…However, compounds 1 and 3 exhibited a small α value (α K1/K3 = 1.29) using n-hexane/EtOAc/ MeOH/water (2:5:2:5, v/v). Subsequently, modification of the solvent system by adding acid, a widely used strategy for HSCCC separation [22,25], was performed by adding acetic acid (0.1%, v/v) to the solvent system n-hexane/EtOAc/MeOH/water (2:5:2:5, v/v). However, this did not improve the α value of compounds 1 and 3 (α K1/K3 = 1.27).…”

“…However, high‐speed countercurrent chromatography (HSCCC) can achieve this goal. As a liquid–liquid partition chromatography, HSCCC adopts a liquid stationary phase (SP) without solid support, which avoids irreversible adsorption and total sample recovery [14, 15]. In addition, load capacity of HSCCC is large.…”
