Resveratrol induces apoptosis of pancreatic cancers cells by inhibiting miR-21 regulation of BCL-2 expression

Abstract: In this study, we demonstrated that the effect of resveratrol on apoptosis is due to inhibiting miR-21 regulation of BCL-2 expression.

“…68,69 In PDAC, BCL2 is overexpressed through the effects of miR-21, which leads to an enhanced anti-apoptotic effect. 70,71 BCL2 is targeted by miR-181b, and the inhibition of BCL2 expression following miR-181b transfection reduces gemcitabine resistance. 72 Programmed cell death 4 (PDCD4) is a tumour suppressor that regulates multiple proteins involved in tumour progression, cell cycle control and differentiation and may be regulated by miR-21 in PDAC, miRNAs in PDACs K Yonemori et al potentially leading to cell death and poor prognosis.…”

“…Functional studies have revealed that miR-21 is an oncogene, which is important in enhancing cell proliferation, promoting cell cycle progression and increasing anti-apoptotic activation in cancer cells (12). In numerous cancer cell lines, miR-21 was demonstrated to be the therapeutic target of resveratrol (13,14). Of note, miR-21 was significantly upregulated in advanced bladder cancer tissues and the overexpression of miR-21 promoted the proliferation of bladder cancer cell lines (15).…”

“…On the other hand, it was illustrated that resveratrol downregulates miR-21 in pancreatic cancer cell lines such as PANC-1, CFPAC-1, and MIA Paca-2. It causes inhibition of Bcl-2 expression; hence, the upregulation of miR-21 modulates Bcl-2 expression and induced apoptosis (Liu et al 2013). Vislovukh et al (2013) noted that resveratrol • Downregulated miR-221 and miR-222 Chen et al (2011) upregulated two miRNAs in MCF-7 breast adenocarcinoma cells namely miR-663 and miR-774.…”
