Resveratrol enhances brown adipocyte formation and function by activating AMP‐activated protein kinase (AMPK) α1 in mice fed high‐fat diet

Abstract: Scope Enhancing the formation and function of brown adipose tissue (BAT) increases thermogenesis and hence reduces obesity. Thus, we investigate the effects of resveratrol (Resv) on brown adipocyte formation and function in mouse interscapular BAT (iBAT). Methods and results CD1 mice and stromal vascular cells (SVCs) isolated from iBAT were treated with Resv. Expression of brown adipogenic and thermogenic markers, and involvement of AMP-activated protein kinase (AMPK)α1 were assessed. In vivo, Resv enhanced … Show more

“…The differentiation and thermogenesis of classical brown and inducible beige/brite adipocytes are controlled by transcriptional regulators and signaling pathways . AMPK, an energy sensor that maintain energy homeostasis, is involved in the polyphenols‐mediated activation of brown fat and browning of WAT . Consistently, in this study, APs promote BAT activity and IngWAT browning in vivo through, at least partly, activation of AMPK α .…”

“…RSV exposure promotes mitochondrial activity in white and brown adipocyte cell models, and both WAT browning and activation of BAT thermogenesis have been implicated in body fat loss after RSV supplementation to adult rodents. In our study, early postnatal RSV supplementation favored the expression of brown adipocyte marker genes in iWAT of adult male mice but not in their BAT, where if anything there was decreased expression.…”

“…More recently, Wang et al . observed a decrease in the average daily weight gain and morphological changes in interscapular BAT (iBAT) in mice treated with resveratrol (0.1% w/w in a high‐fat diet) for 4 weeks.…”

“…Furthermore, the authors observed an increase, both in UCP1 gene expression and in protein expression of PRDM16 and cytochrome c , master factors involved in brown adipocyte formation, in the resveratrol‐ supplemented animals. They concluded that the enhanced formation and function of iBAT in mice fed the high‐fat diet and treated with resveratrol were associated with AMPK activation …”
