Resveratrol and insulin association reduced alveolar bone loss and produced an antioxidant effect in diabetic rats

Abstract: Background: The present investigation studied the effects of systemic administration of resveratrol (RSV) on the development of experimental periodontitis (EP) and on the release of markers of inflammation, bone metabolism, and oxidative stress in diabetic rats. Methods: Seventy-five male rats were divided into five groups: DM+PLAC: Diabetes Mellitus + placebo solution; DM+INS: DM + insulin therapy; DM+RSV: DM + RSV; DM+RSV+INS: DM + RSV and insulin; NDM: non-diabetic. Streptozotocin was used to induce DM and … Show more

“…The therapeutic potential of RSV for DP has been explored in three animal studies and two clinical trials, as tabulated in Table 2. All investigations agreed on the anti-inflammatory effects of RSV [18,20,21,76], and three confirmed its anti-diabetic [18,20,21] capabilities for managing DP. The results of these studies revealed a remarkable attenuation of bone loss, reduced inflammatory cytokines, and improved fasting blood glucose levels, so it can be considered an effective approach for reducing bone resorption and improving metabolic management.…”

“…1.5 × 10 -3 % hydrogen peroxide solution can decrease the survival rate of gingival fibroblasts and promote the secretion of TNF-α by gingival fibroblasts, which can be inhibited by non-enzymatic antioxidant α-tocopherol (vitamin E) [23]. The combination of resveratrol and insulin can reduce the level of pro-inflammatory factors and increase the level of SOD, thereby inhibiting the progress of experimental periodontitis, and reducing the loss of alveolar bone, which has a beneficial effect on the repair of bone tissue [24]. Some scholars collected serum and gingival specimens by local injection of vitamin C into the subgingival periosteum of male SD rats [25].…”

“…Antioxidant compounds, such as resveratrol, quercetin, and curcumin, are increasingly proposed to rebalance the oxidative stress-loaded environment at the ground of multiple diseases (Sczepanik et al 2020). In an experimental periodontitis model in diabetic rats, systemic resveratrol reduced NADPH-oxidase levels in the gingiva, upregulated mRNA levels for osteoprotegerin, and reduced the progression of periodontitis independently from insulin treatment (Cirano et al 2020). Quercetin is a flavonoid, which has potent anti-inflammatory, antibacterial, and antioxidant effects.…”
