The change in strain-specific urine odors which appears after bone marrow transplantations was systematically examined in mice in order to demonstrate an influence of the hematopoietic system on urinary chemosignals. Four mice were trained in a Y maze to discriminate two fully allogeneic mouse strains via their urine odors. Urine samples obtained from three inbred strains, from syngeneic reconstituted mice, and from allogeneic reconstituted chimeras were combined in a number of ‘transfer of training’ tests. The strain-specific urine odors of the recipients were changed by a fully allogeneic bone marrow transplant. Since this change could not be found in syngeneic reconstituted mice, we concluded that it was caused by the graft. Experiments partly failed to demonstrate donor- and recipient-specific components in the urine odor of the chimeras.

In a rat model we describe the role T lymphocytes play in the immunogenicity of bone marrow. Mainly they modulate the MLC-like stimulatory cell potential and the MHC class II antigen expression of nonlymphoid bone marrow cells via the release of newly detected specific soluble factors and immunologically unspecifîc ones. These findings are discussed in relation to the graft rejection encountered in clinical bone marrow transplantation following certain T depletion protocols.

Absorption procedures which allow the production of a selectively cytotoxic anti-human lymphocyte serum are described. Although the production of a reagent whose reactivity is restricted. Although the production of a reagent whose reactivity is restricted exclusively to lymphocytes may be achieved by exhaustive absorption steps using fresh human erythrocytes, CML cells, and fetal liver cells, a more realistic alternative is the use of appropriately selected cultured human leukemia cell lines. Data are presented which show how these cell lines may be employed to selectively manipulate the cross-reactivity spectrum of ALS. Pre-treatment of donor bone marrow cells prior to transplantation with a selectively lymphocytotoxic ALS has been shown to allow transplantation of bone marrow across major histocompatibility barriers in rodents without the occurrence of GvH reactions, and it is the purpose of the present investigations to show that an analogous anti-human ALS can be prepared which possesses the required degree of selectivity to allow its application for human bone marrow transplantation.
