Resistome characterization of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli isolated from wastewater treatment utilities in Oregon

Abstract: Infections resistant to broad spectrum antibiotics due to the emergence of extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae is of global concern. This study characterizes the resistome (i.e., entire ecology of resistance determinants) of 11 ESBL-producing Escherichia coli isolates collected from eight wastewater treatment utilities across Oregon. Whole genome sequencing was performed to identify the most abundant antibiotic resistance genes including ESBL-associated genes, virulence factors… Show more

“…Bacteria carrying extended-spectrum beta-lactamases (ESBL) commonly possess one of the three gene types bla TEM , bla CTX-M , and bla SHV , thereby enhancing their ability to develop resistance against β-lactam antibiotics [ 38 ]. Moreover, ESBL-producing E. coli are more inclined to exhibit resistance to multiple drugs than E. coli that do not produce ESBL [ 39 ]. In the present study, the majority of MDR E. coli strains possessed bla TEM and bla CTX-M , with the presence of bla SHV notably lacking.…”

“…E. coli strains were previously isolated from influent, secondary effluent, final effluent, and biosolids streams of eight wastewater treatment utilities across Oregon as described in our previous study (Table S1) (26). Wastewater samples were collected between January 2019 and September 2020.…”

“…Long-read sequencing libraries were prepared using the Rapid Barcoding Sequencing kit (SQK-RBK004) and sequenced on a MinION system (Oxford Nanopore, Oxford, UK) following the manufacturer's protocol. Short-read sequencing was performed using the Illumina MiSeq as described previously (26).…”
