Determination of resistance to the attack of Zabrotes subfasciatus (Coleoptera: Bruchidae) in bean genotypesZabrotes subfasciatus (Boheman, 1833) (Coleoptera: Bruchidae) is considered the main stored bean pest. The most widely used method of control is the use of insecticides. However, other control methods may be applied, such as plant resistance. Aiming to find alternative strategies to its control, this study evaluated the types of resistance in 12 bean genotypes. The experiment was carried out at the Laboratory of Plant Resistance to Insects of the Department of Phytopathology of the FCAV/UNESP, Jaboticabal/SP, from March to November 2009. Oviposition non-preference (free choice and no-choice) tests and the no-choice test for feeding were carried out. The results demonstrated that: the genotypes Dor 391, Dor 476, IAPAR-MD 806, and Raz 59 showed non-preference to oviposition by Z. subfasciatus, in the free-choice test; in the no-choice test, all genotypes were equally oviposited by the insect; in the free-choice chance test, males and females were equally attracted by all tested genotypes and Raz 56, Arc 2, Raz 55, Raz 49, and Raz 59 showed non-preference to feeding and/or antibiosis.Key words: Phaseolus vulgaris L., stored bean, host plant resistance, Mexican bean weevil. O Zabrotes subfasciatus (Boheman, 1833) (Coleoptera: Bruchidae) é considerado a principal praga do feijão armazenado. O método de controle mais utilizado tem sido o uso de inseticidas; no entanto outros métodos podem ser empregados, como, por exemplo, a resistência de plantas. Este trabalho teve por objetivo determinar os tipos de resistência em 12 genótipos de feijoeiro ao ataque dessa praga. O experimento foi conduzido no Laboratório de Resistência de Plantas a Insetos, do Departamento de Fitossanidade da FCAV\UNESP, Jaboticabal -SP, no período de março a novembro de 2009. Foram realizados testes de não preferência para oviposição, com e sem chance de escolha, e de alimentação, sem chance de escolha. Concluiu-se que os genótipos Dor 391, Dor 476, IAPAR-MD 806 e Raz 59 apresentam resistência do tipo não preferência para oviposição por Z. subfasciatus, em teste com chance de escolha; no teste sem chance de escolha todos os genótipos foram igualmente ovipositados pelo inseto; no teste com chance de escolha, machos e fêmeas foram igualmente atraídos por todos os genótipos testados; e Raz 56, Arc 2, Raz 55, Raz 49 e Raz 59 apresentaram resistência dos tipos não preferência para alimentação e,ou, antibiose. Resumo -Na busca de alternativas ao controle químico do caruncho Zabrotes subfasciatus (Coleoptera:Bruchidae), avaliou-se, em feijoeiro (Phaseolus vulgaris L.), o efeito de três genótipos, cv. Carioca e duas linhagens contendo a proteína arcelina (Arc1 e Arc3), e da adição nos grãos armazenados de óleo de soja (Glycine max Merrill); óleo de nim (Azadirachta indica A. Juss.), munha (resíduo de trilha da colheita), calcário dolomítico e terra de formigueiro, comparativamente aos grãos não tratados e ao controle químico com malathion 500 CE. O experimento foi realizado no laboratório da Embrapa-Centro Nacional de Pesquisa de Arroz e Feijão, localizado no Município de Santo Antônio de Goiás, GO, em condições não controladas. Na linhagem Arc1, constatou-se maior proteção aos danos do caruncho, observando-se redução do número de ovos e de adultos emergidos e da porcentagem de sementes danificadas em relação à Arc3 e à cv. Carioca. Quando os grãos foram tratados com malathion, óleo de nim e óleo de soja, observou-se menor número de ovos e de adultos emergidos e redução de danos, e não houve diferença na porcentagem de sementes danificadas entre o malathion e o óleo de nim. A mistura das sementes com terra de formigueiro conferiu baixa proteção ao caruncho, enquanto os tratamentos com munha e calcário dolomítico não apresentaram eficiência na redução da progênie e dos danos de Z. subfasciatus.Termos para indexação: Phaseolus vulgaris, controle de pragas, resistência aos pesticidas. The experiment was carried in a laboratory at Embrapa-Centro Nacional de Pesquisa de Arroz e Feijão, Santo Antônio de Goiás, GO, Brazil, under noncontrolled conditions. The highest levels of protection against Z. subfasciatus were observed in Arc1, with reduction in the number of eggs, number of emerged adults and percentage of damaged seeds than in Arc3 and Carioca cultivar. The number of eggs, number of emerged adults and the percentage of damaged seeds were reduced when the seeds were treated with malathion, neem and soybean oils. There was not difference in percentage of damaged seeds between malathion and neem oil. The treatment of the seeds using ant hill ground showed low efficiency, and trashing residues and dolomitic lime did not show reduction on the progeny and damage of Z. subfasciatus. Arcelin, a seed protein originally discovered in wild bean accessions, was purified, characterized, and compared to phaseolin, the major seed protein of common bean, and to phytohemagglutinin (PHA), the major bean seed lectin. Arcelin and PHA has several characteristics in common.Both were glycoproteins having similar subunit M, deglycosylated Mn and amino acid compositions. The two proteins were related antigenically and they had the same developmental timing of accumulation. Arcelin also had some hemagglutinating activity, a characteristic associated with lectins. However, several features distinguished arcelin from PHA. Arcelin had a more basic isoelectric point than PHA, greater numbers of basic amino acid residues, additional cysteine residues, and one methionine residue, which PHA lacks. Native PHA protein is a tetramer of subunits, and although a small component of native arcelin protein was also tetrameric, most of the arcelin preparation was dimeric. The hemagglutinating activity ofarcelin was specific only for some pronase-treated erythrocytes. It did not agglutinate native erythrocytes, nor did it bind to thyroglobulin or fetuin aff'mity resins as did PHA. Although arcelin has lectin-like properties, we believe the distinctions between arcelin and PHA warrant the designation of arcelin as a unique bean seed protein.was co-dominant with respect to each other and dominant with respect to alleles for the absence of arcelin. In this study, we report on the purification and characteriz,tion of the arcelin-1 variant and compare its properties to those of PHA and phaseolin seed proteins. The major seed storage proteins of cultivated common bean (Phaseolus vulgaris) are phaseolin and PHA,2 also referred to as bean lectin. In several wild bean accessions, a third important protein fraction has been observed which had not been described previously in bean cultivars (19,23). This fraction was discovered as a major seed protein having a different electrophoretic mobility than either phaseolin or PHA with which it occurred in seeds of some wild beans. Four electrophoretic variants of this protein were observed which consisted of distinct groups of polypeptides having similar mobilities on two-dimensional gels.
