Resistance to quaternary ammonium compounds in Staphylococcus spp. isolated from the food industry and nucleotide sequence of the resistance plasmid pST827

Abstract: The complete nucleotide sequence of the 2.8 kb plasmid pST827 involved in resistance to the quaternary ammonium compound (QAC) benzalkonium chloride in meat-associated staphylococci was determined. An open reading frame (ORF) similar to the QAC resistance genes qacC, ebr and smr previously reported from clinical staphylococcal strains was identified (qacC'). In addition an ORF coding for a protein (Rep827) showing extensive homology to reported replication proteins of Gram-positive organisms was found. The occ… Show more

“…PCR amplification with smr-specific primers generated amplicons of 285 bp with template DNA from the isolate that harbored the 2.2-kb plasmid; the PCR result was negative for the isolate that did not contain the 2.2-kb plasmid. The nucleotide sequence of the PCR product was found to be 99% homologous to the DNA sequence of the smr (qacCЈ) gene on pST827 published previously (6). None of the 23 penicillin-and tetracycline-susceptible S. aureus isolates contained plasmids.…”

“…1). The Pribnow box TAAAAT is identical to the corresponding sequence in plasmid pST827 (6). Upstream of the smr gene, transcribed in the opposite direction, is a 999-bp sequence (nt 31 to 1030) representing an ORF, designated repNVH99, that encodes a replication protein.…”

“…aeruginosa, and have suggested that the increases in quantities of cellular membrane components, such as phospholipids, fatty acids and neutral lipids, suppress the adsorption of benzalkonium chloride molecules to the cell. Apart from such an alteration in the cell envelope, the active efflux of cationic surfactant, as an alternative mode of the acquired resistance, has also been shown in staphylococci (Littlejohn et al 1992;Heir et al 1995Heir et al , 1998. These two distinct modes of acquired resistance have often been described for antibiotics and, in particular, the increased development of an efflux system brings about multi-drug resistance (Nikaido 1994).…”

Generally, it is important to take measures against the emergence of Gram-negative bacteria, which are resistant to biocides or antiseptics in nature due to their cell surface structure, compared with Gram-positive bacteria (Denyer and Maillard, 2002).…”
