Requirement of a bone morphogenetic protein for the maintenance and stimulation of osteoblast differentiation

Abstract: The requirement of a bone morphogenetic protein for the maintenance and stimulation of an osteoblast phenotype was examined using mouse MC3T3-E1 cell cultures. Cells expressed BMP-4 mRNA, which correlated with the stimulation of the osteoblast phenotype. The addition of a BMP-4 specific antibody reduced bone nodules, suggesting that BMP-4 is required for the osteogenic activity of osteoblasts in an autocrine manner. Exogenously added BMP-7 gradually decreased the expression of BMP-4 with a concurrent stimulati… Show more

“…In fact, the expression of both the BMP-2 mRNA and protein were enhanced greatly after three days of culture in this study, which implies the induction of osteoblast differentiation. However, the decrease after seven days of culture indicates that a negative feedback mechanism might act against its overexpression after three days of culture 23) . Integrins are heterodimers consisting of subunits comprising α and β chains, and exist in the cell membrane.…”

“…MC3T3-E1 cells synthesize BMP-4, which induces osteoblast differentiation (Martinovic et al, 2006;Wang et al, 2006). We next examined the effect of an antibody against BMP-4 on the mineralization of MC3T3-E1 cells induced by CS-E.…”

“…They were discovered and named based on their osteoinductive ability. Today we know that BMPs have an essential role in embryonic development and tissue differentiation, regulation of cell survival, and apoptosis, as well as in the organogenesis of skeletal and nonskeletal tissues (14,45,60,67,68,69,74).…”
