Aims: Depression is a mental health issue that starts most often in early adulthood and it is a common and recurrent disorder causing significant morbidity and mortality worldwide. Amitriptyline is a tricyclic antidepressant that is known to inhibit the presynaptic reuptake of serotonin, norepinephrine, and inhibitor of mitochondrial functions and induces apoptosis in several tissues. This study aims to identify the changes in liver and kidney structure and functions after treatment of male rats with Amitriptyline drugs. Materials and Methods: A total of 20 male albino rats were randomly and equally divided into 2 groups (G1, control group that included animals that did not receive any treatment during the experimental period. G2, Amitriptyline (Tryptizol; El Kahira Pharm And Chem Ind Co) group in which rats were injected intraperitoneally with Amitriptyline (100 mg/kg body weight/daily) for four weeks). Results: The current results revealed that; Amitriptyline treatments significantly (P <0.05) increased the levels of serum ALT, AST, ALP, urea, creatinine, sodium ions, chloride ions and liver and kidney damages as compared to control. In contrast; a significant (P <0.05) decrease in albumin, and total protein, potassium ions and calcium ions in Amitriptyline group was reported when compared with control group. Conclusion: Amitriptyline has many side effects on rat liver and kidney, it induced liver and kidney toxicity and tissue injury were it metabolized to nortriptyline which inhibits the reuptake of norepinephrine and serotonin almost equally. Amitriptyline inhibits the membrane pump mechanism responsible for uptake of norepinephrine and serotonin in adrenergic and serotonergic neurons.

Effect of L-carnitine on potassium dichromate-induced nephrotoxicity in rats: modulation of PI3K/AKT signaling pathway

Background and purpose: Kidney diseases impose significant global health challenges. Potassium dichromate (PD) is a heavy metal frequently associated with nephrotoxicity. PD prompts oxidative and inflammatory injuries in renal tissues. L-carnitine is a naturally-occurring amino acid commonly used as a supplement. Experimental approach: Forty rats were randomly allocated into 5 groups. Group 1 (normal) received only saline. Nephrotoxicity was induced in the remaining groups by PD (15 mg/kg; i.p). Group 2 served as a nephrotoxic group. Groups 3-5 received L-carnitine (25, 50, and 100 mg/kg; p.o.), respectively for 4 weeks. Findings/Results: PD administration resulted in elevated serum creatinine and blood urea nitrogen accompanied by diminished reduced glutathione and elevated malondialdehyde, tumor necrosis factor-alpha, and transforming growth factor-beta renal tissue contents relative to normal rats. PD also produced apoptotic histopathological injuries and down-regulated PI3K/Akt signaling pathway; signifying ongoing apoptosis. In the current work, L-carnitine use in the selected dose levels resulted in improvement of all the aforementioned serum, renal tissue, and histological parameters relative to nephrotoxic rats. L-carnitine up-regulated PI3K/Akt signaling pathway that was down-regulated post PD use. Conclusion and implications: Collectively, the study highlighted that the possible mechanisms beyond the beneficial effects of L-carnitine are mainly through its antioxidant as well as anti-inflammatory actions. L- carnitine significantly abrogated apoptosis via up-regulation of PI3K/Akt signaling pathway and signified restoration of normal renal cell proliferation and functionality.

This study was designed to investigate the changes in total oxidant (TOS)/antioxidant (TAS) and oxidative stress index (OSI) levels in liver and kidney tissues of mice pre-treatment of artemisinin against oxidative stress that may occur in mice administered pentylenetetrazole (PTZ). Swiss albino mice (Male) (n=42) were used in the study. The mice were divided into six groups and each group had seven animals (n=7): (1) Control (C) /saline Group, (2) PTZ (35 mg/kg) Group, (3) Valproate (VPA) (100 mg/kg)+PTZ Group, (4) Artemisinin (ART) (30 mg/kg)+PTZ Group, (5) ART (60 mg/kg)+PTZ Group, (6) ART (120 mg/kg)+PTZ Grubu. Mice received injections intraperitoneally (i.p.). After the treatments, the animals were observed for seizures for 30 minutes. On the last day (day 26) of the experiment, the PTZ loading dose (75 mg/kg) was administered to the mice and then the animals were sacrificed. TAS, TOS and OSI levels were measured in liver and kidney tissue. PTZ increased TOS and decreased TAS in liver and kidney tissue.
