Remapping of the stripe rust resistance gene Yr10 in common wheat

Abstract: Yr10 is an important gene to control wheat stripe rust, and the search for Yr10 needs to be continued. Wheat stripe rust or yellow rust is a devastating fungal disease caused by Puccinia striiformis f. sp. tritici (Pst). Host disease resistance offers a primary source for controlling wheat stripe rust. The stripe rust resistance gene Yr10 confers the race-specific resistance to most tested Pst races in China including CYR29. Early studies proposed that Yr10 was a nucleotide-binding site, leucine-rich repeat ge… Show more

“…The maize Ubiquitin ( Ubi ) promoter (Christensen et al ., ) is a constitutive promoter widely used in research. The wheat Wks1 promoter (Fu et al ., ), on the other hand, is an alternative promoter that has been successfully used to express transgenes (Yuan et al ., ). All of the remaining constructs derived from the ICS and PAL genes were based on the Ubi promoter.…”

“…The identification of underlying genes with annotations related to the trait provides further reliability for the MTAs identified. For instance, four genes (TraesCS4D01G096900: Lr, TraesCS7A01G517300: Lr, TraesCS4B01G007000: Sr, and TraesCS3A01G495400: Lr) were annotated as the leucine-rich repeat protein family, which was previously reported to have a potential role in Yr [38,39] and Sr resistance [40]. Similarly, five genes (TraesCS2A01G076800: Yr, TraesCS7B01G473300: Yr, TraesCS1A01G325300: Sr, TraesCS6B01G394600: Yr, and TraesCS5B01G199700: Sr) were annotated as the F-box family protein, which were reported to have potential role in Lr [41], Sr [42], and Yr [43] resistance in wheat.…”

“…Maize ubiquitin ( Ubi-1 ) promoter is contemplated as a better choice compared to CaMV 35S and Adh1 for the constitutive expression of a transgene, particularly in monocots [ 29 ]. It has extensively been used for the transient as well as stable expression of transgenes in several monocots including wheat [ 30 ], maize [ 31 ], rice [ 32 ], sugarcane [ 33 ], and sorghum [ 34 ]. SUGARWIN1 and SUGARWIN2 genes were cloned in pUbiAB vector under maize ubiquitin promoter ( Ubi-1 ) and nos terminator.…”
