Reliability of Transcutaneous Measurement of Renal Function in Various Strains of Conscious Mice

Abstract: Measuring renal function in laboratory animals using blood and/or urine sampling is not only labor-intensive but puts also a strain on the animal. Several approaches for fluorescence based transcutaneous measurement of the glomerular filtration rate (GFR) in laboratory animals have been developed. They allow the measurement of GFR based on the elimination kinetics of fluorescent exogenous markers. None of the studies dealt with the reproducibility of the measurements in the same animals. Therefore, the reprodu… Show more

“…Therefore, sequential measurements within the same animal have been limited by the welfare of the animals and methodological reasons. The proved withinanimal reproducibility of the transcutaneous measurement 13 makes it an ideal method for detecting changes in kidney function due to disease progression or ageing. For the same reason, it can also be extremely useful to study the effectiveness of therapeutic approaches.…”

“…This could attemper the consequences of the above altered vascular tone. Moreover, a reduced left ventricular ejection fraction [21] and a normal renal function [25] reported in SV129 mice could also limit the consequence of eventually increased peripheral vascular resistances on BP.…”

“…A miniaturized Non-Invasive-Clearance (NIC) technique to measure kidney function in conscious mice was applied to determine GFR, as previously described [16, 17]. The NIC-Kidney device (Mannhein Pharma and Diagnostics, Mannheim, Germany) allows for transcutaneous measurement of the elimination kinetics of the fluorescent renal marker FITC-S without surgery or blood and urine collection.…”
