Relationship between PGCs Settle and Gonad Development in the Early Chicken Embryo

Abstract: Chick embryos from stage 14 to stage 31 were studied by means of serial section and light microscopy in order to learn the relationship between the settlement sites of the primordial germ cells (PGCs) and the forming genital ridge. The results showed that: when embryo hatched for 53-56 h, the PGCs reached the coelomic epithelial tissue where gonad would be formed, meanwhile the epithelial tissue began thicker before the PGCs reached. Before stage 19, the final region the PGCs arrived was the thickened portion … Show more

“…We next further explored the specific function of Notch signaling during germ cell formation. To this end, we detected the formation of PGCs (4.5 days) [ 20 ] and SSCs (18 days) [ 21 , 22 ] after activation and inhibition of Notch signaling during chick embryo hatching. With respect to PGCs, we found that the activation of Notch signaling significantly upregulated the expression of Lin28 (8.3427 ± 0.23, p < 0.05) and Blimp1 (12.4213 ± 0.13, p < 0.05), which mark formation of PGCs at 4.5 days.…”

“…The origin, migration, accumulation and settlement process of reproductive cells are well established (Li et al . ). Chickens also possess a unique advantage in their migration patterns, which make germ cells easily separable during early development.…”

“…A total of 18,340 eggs were used in this experiment, which were divided into three groups as three biological replicates. The eggs used for collection of embryonic stem cells (ESC) were used immediately, while those used for isolation of primordial germ cell (PGC) or isolation of spermatogonial stem cell (SSC) were incubated at 37°C and 75% relative humidity for 5.5 and 19 days, respectively [ 11 ].…”
