Relationship Between Marek's Disease and the Time Course of Viral Genome Proliferation in Feather Tips

Abstract: A total of 114 male chickens from three sire families of a commercial cross of White Leghorn chickens were infected with RB-1B Marek's disease (MD) virus at 21 days of age by exposing them to chickens previously inoculated with MD virus. The presence of virus in feather tips, feather pulp, and MD viral antibodies indicated all chickens became infected. The first virus-positive chickens were observed at 12 days postexposure (dpe). The frequency reached a maximum at 27 dpe and then decreased. At 80 dpe, when the… Show more

“…Samples of the lung and spleen were collected directly into cell lysis solution (PURAGENE † DNA isolation kit; Gentra Systems, Inc. Minneapolis, Minnesota, USA). Feathers with fleshy calamus were collected from the alar tracts and FP was extracted as reported earlier (Kuhnlein et al, 2006). Real-time polymerase chain reaction (PCR) assay was performed as described previously (Gimeno et al, 2008).…”

“…Feather pulp (FP) from four feathers with fleshy calamus was collected from the alar tracts. FP was extracted as reported earlier (Kuhnlein et al, 2006). Briefly, pulp was squeezed from the calamus with a sterile pipette tip and added to the cell lysis solution.…”

“…Extraction of the DNA from feather tips was carried out using a protocol adapted from Kuhnlein et al (2006). Feather tips were cut into small pieces with sterile scissors and placed into 1.5-mL tubes containing 400 L of extraction buffer (2% 2-mercaptoethanol, 10 mM Tris-HCL at pH 8.0, 100 mM NaCl, 10 mM EDTA at pH 8.0, and 0.5% SDS).…”
