Relation of sodium pump inhibition to positive inotropy at low concentrations of ouabain in rat heart muscle.

Abstract: SUMMARY1. Low concentrations of ouabain which produce a positive inotropic effect on rat ventricular muscle do not inhibit the isolated Na+-K+-ATPase enzyme from this tissue, suggesting that these low-concentration inotropic effects are not related to sodium pump inhibition (Erdmann, Philipp & Scholz, 1980; Adams, Schwartz, Grupp, Grupp, Lee, Wallick, Powell, Twist & Gathiram, 1982).2. We tested this hypothesis by continuously measuring intracellular Na+ activity with Na+-selective micro-electrodes and, separ… Show more

“…The steady state level of aNa in guinea-pig papillary muscle continuously stimulated at a frequency of 0.1 Hz was 7.8 + 2.0 mi. This value compares favorably with mean values reported for aNa: measurements of aNa include 5.7-8.5 mM in rabbit and guinea-pig ventricular muscles (LEE and FOZZARD, 1975;KLEBER, 1983;Grupp et al, 1985). In cardiac Purkinje fibers COHEN et al (1982) showed that aNa increased dependent on stimulation frequency and after cessation of stimulation the elevated aNa declined in an exponential manner.…”

“…), respectively. Both the values for aK and aNa were within the range reported for ventricular muscles using single-barrel ion-selective microelectrodes: for aK, 82.6-116 mM (LEE and FOZZARD, 1975;BROWNING and STRAUSS,1981;COHEN et al, 1982); and for aNa, 5.7-8.5 mM (LEE and FOZZARD, 1975;COHEN et al, 1982;KLEBER, 1983;GRUPP et al, 1985).…”

“…Ouabain has been thought to increase the contraction through a Na+-Ca2+ exchange mechanism (Reuter & Seitz, 1968;Sheu & Fozzard, 1982;Wasserstrom et al, 1983;Eisner et al, 1984;Vassalle & Lee, 1984;Wier & Hess, 1984;Grupp et al, 1985). Ouabain inhibits Na'-K' ATPase and results in an accumulation of [NaJ]i.…”

“…Much of our present knowledge of cellular Na+ regulation has emerged from studies utilizing Na+-sensitive microelectrodes which allow intracellular Na+ activity (aNa) to be monitored continuously over time and sensitively enough to detect even small changes in ion activity. Na+-sensitive microelectrodes have proved to be a powerful tool in elucidating the mechanism of action of cardiac steroids (Lee, 1985) and particularly in resolving the controversial issue on the positive inotropic effect of low ouabain concentrations in rat heart (Grupp, Im, Lee, Lee, Pecker & Schwartz, 1985).…”
