Changes in intra- and extracellular potassium and intracellular sodium activities induced by repetitive stimulation and their relation to membrane potential in guinea-pig papillary muscle.

Abstract: We measured changes in membrane potential (MP), extra-and intracellular potassium and intracellular sodium ion activities (4, 4, and aNa) in papillary muscle of the guinea-pig heart induced by repetitive stimulation at various frequencies and periods in vitro, using doublebarrelled ion-selective microelectrodes. Stimulation (2-4 Hz) depolarized the cells, and termination of stimulation induced hyperpolarization. The a K and aNa (stimulated at 0.2 Hz) were 92.3 + 4.6 and 7.8 ± 2.0 mM (mean + S.D.), respectively… Show more

“…The method of preparing K+-selective microelectrodes (KISEs) was essentially the same as that described by HOTOKEBUCHI et al (1987). In brief, two capillaries (Narishige, GD-1) were mounted in parallel on a horizontal micropipette puller (Narishige, PW-77), and heat was applied.…”

“…The DC-resistance of the K+-selective barrel and the reference barrel were 10-100 GSA and 20-60 MS2, respectively. The KISE, thus prepared, was stable for more than 12 h. The "response time" (HOTOKEBUCHI et al, 1987) of the Vol. 39, No.…”

“…The methods used to fabricate the ion-sensitive, double-barreled electrodes were reported previously [16], and are described briefly here. Two glass capillary tubes with a glass fiber inside were mounted in parallel on a horizontal micropippet puller (PD-5; Narishige, Tokyo, Japan).…”
