Regulation of human natural killing by levamisole

Abstract: The effect of levamisole on human natural killing (NK) has been studied. In short-term chromium release assays, levamisole at a concentration of 10(-3) M was inhibitory to NK when present in the assays. Pretreatment of NK effector cells and K562 target cells with levamisole separately indicated that the effect was on effector cell activity and was not due to any change in target cell susceptibility. Inactivation of the effector cells required greater than 4 h pretreatment with levamisole if NK activity was sub… Show more

“…The correlation of granule content and cytolytic capacity within NK cells has been well documented (21)(22)(23)(24)(25)(26)(27). However, it is unclear how much degranulation is sufficient for lysis and, for that matter, to what extent other nongranular mechanisms contribute.…”

“…There is no evidence that levamisole can augment NK cells [24]. In vitro investiga tions in our study on RCC patients showed that levami sole and OMPI are apparently without any influence on NK cell activity.…”

“…Preparation ofperipheral blood lymphocytes (PBL) Human peripheral blood mononuclear cells were isolated from defibrinated or heparinized blood as previously described (Shau & Dawson, 1982). Adherent cells were depleted by incubating the mononuclear preparations (4 x 106 cells ml-1) at 370 for 1 h on plastic tissue culture flasks and by passing cell suspensions through nylon wool columns.…”
