The aim of the present study was to identify the gene expression profile of three candidate genes namely heat shock protein 70, oviductal glycoprotein and osteopontin in buffalo oviduct during different stages of the oestrous cycle. Slaughter house derived buffalo oviducts were categorized as belonging to early, mid and late phase of the oestrous cycle based on the morphological appearance of CL and progesterone concentration. The relative abundance of each of the candidate gene transcript was quantified by real time RT-PCR. Transcripts of all the three candidate genes were found to be expressed in OEC throughout the oestrous cycle. The results of the present study demonstrate that gene expression in the buffalo oviduct is clearly regulated during the oestrous cycle, as these candidate genes play a crucial role in several reproductive processes. It was found that Hsp 70 gene was significantly upregulated in OEC belonging to the early and late phases when compared to mid phases of the cycle. The expression of OVGP mRNA was found to be maximum during the early phase and it remained low during the mid and late phases while expression of OPN mRNA was maximum during the early phase and declined sequentially during the mid and late phase of oestrus cycle.

In cattle, pro-oestrous oestradiol and dioestrous progesterone concentrations modulate endometrial gene expression and fertility. The aim was to compare the effects of different periovulatory endocrine profiles on the expression of progesterone receptor (PGR), oestrogen receptor 2 (ESR2), oxytocin receptor (OXTR), member C4 of aldo-keto reductase family 1 (AKR1C4), lipoprotein lipase (LPL), solute carrier family 2, member 1 (SLC2A1) and serpin peptidase inhibitor, clade A member 14 (SERPINA14): (1) between uterine horns ipsi- and contralateral to the corpus luteum (CL), (2) between regions of the ipsilateral horn and (3) in the vagina. Endometrium and vagina tissue samples were collected from cows that ovulated a larger (large follicle-large CL, LF-LCL; n=6) or smaller follicle (small follicle-small CL, SF-SCL; n=6) 7 days after oestrus. Cows in the LF-LCL group had a greater abundance of transcripts encoding ESR2, AKR1C4, LPL, SLC2A1 and SERPINA14, but a reduced expression of PGR and OXTR in the endometrium versus the SF-SCL group (PPGR and OXTR was greater in the contralateral compared with the ipsilateral horn (PPGR, ESR2, LPL, SLC2A1 and SERPINA14 (P<0.05). Different periovulatory endocrine profiles, i.e. LF-LCL or SF-SCL, did not influence gene expression in the vagina and had no interaction with inter- or intra-uterine horn gene expression. In conclusion, inter- and intra-uterine horn variations in gene expression indicate that the expression of specific genes in the bovine reproductive tract is location dependent. However, spatial distribution of transcripts was not influenced by distinct periovulatory sex-steroid environments.

Objective: To evaluate the effect of excess fat and suckling control on postpartum reproductive activity of hair ewes. Design/Methodology/Approach: Eighty-one (81) hair ewes with average weight of 58±8 kg were used, which on postpartum day 7 were assigned to one of four treatments under a completely randomized design with 2×2 factorial arrangement. Factor A is the type of suckling [continuous suckling (CS) and suckling control (SC)] and factor B, excess fat (with or without). In CS, ewes remained with their offspring 24 hours a day (n=16). In continuous suckling plus fat (CSF), ewes remained with the offspring all day plus the addition of 8% in dietary fat (n=24). In SC, it was 30 min of suckling only twice per day (n=14). In suckling control plus fat (SCF), controlled suckling lasted 30 min, twice a day plus 8% fat (n=25). Results: The response to the treatments was similar (p > 0.05) in onset of estrus, return rate, gestation, prolificacy and fertilization; for the lambing rate variable, there were differences (p ≤ 0.05), the SCF treatment was higher (68%) compared to CS (50%) and CSF (50%), but equal (p < 0.05) to SC (57.1%). Implications: In this experiment, it was observed that the SCF treatment presented a better response, implying that controlling suckling and providing excess fat helps to restore ovarian activity sooner after lambing. Conclusions: Suckling control plus inclusion of dietary excess fat in ovulation induction protocols during early postpartum (around day 25 postpartum) can be included in routine reproductive management of flocks, improving the reproductive efficiency of hair sheep.
