Regional Metabolic Patterns of Abnormal Postoperative Behavioral Performance in Aged Mice Assessed by 1H-NMR Dynamic Mapping Method

Abstract: Postoperative abnormal neurobehavioral performance (PANP) is a common phenomenon in the early postoperative period. The disturbed homeostatic status of metabolites in the brain after anesthesia and surgery might make a significant contribution to PANP. The dynamic changes of metabolites in different brain regions after anesthesia and surgery, as well as the potential association with PANP is still not well understood. Here, a battery of behavioral tests was used to assess the effects of laparotomy under isoflu… Show more

“…The metabolic information, required to determine metabolic fluxes, commonly vary with cerebral regions as shown in the human, 44 rat, 45 and mouse 46 brains. Both the cerebral cortex and the deep brain regions have their own variation characteristics after SCI.…”

“…To minimize the impact of post-mortem changes on cerebral metabolites [ 28 ], the anesthetized rat was microwaved using our previous method [ 29 ]. After euthanasia, the rat brain was quickly removed according to the Allen Brain Atlas [ 30 ] and dissected into 12 different regions (as indicated in Figure 8 ) [ 31 ]. Briefly, the olfactory bulb (OB) and the cerebellum (CE) were first sampled, followed by the hippocampus (HP), the thalamus (THA) and the striatum (STR).…”

“…Recently, our laboratory has used the proton nuclear MRS to investigate specific patterns of spinal metabolites underlying histamine-independent pruritogen α-Me-5-HT-evoked pruritus compared with histamine and capsaicin [7] . Results from these studies provide a general strategy for the previously proposed method [9][10][11][12] . Proton nuclear MRS is a commonly used tool to measure levels of N-acetylaspartate (NAA), glutamate (Glu), creatine (Cr), myo-inositol (Ins), etc.…”
