Region‐specific disturbed iron distribution in early idiopathic Parkinson's disease measured by quantitative susceptibility mapping

Abstract: In Parkinson's disease (PD), iron elevation in specific brain regions as well as selective loss of dopaminergic neurons is a major pathologic feature. A reliable quantitative measure of iron deposition is a potential biomarker for PD and may contribute to the investigation of iron-mediated PD. The primary purpose of this study is to assess iron variations in multiple deep grey matter nuclei in early PD with a novel MRI technique, quantitative susceptibility mapping (QSM). The inter-group differences of suscept… Show more

“…Related studies have found regional susceptibility or R2* differences in single individual anatomical structures such as the putamen, caudate nucleus or the globus pallidus [17,34,35]. Extending these findings, we were able to demonstrate a more widespread involvement of the deep gray matter structures by applying QSM, including the substantia nigra, nucleus ruber, thalamus, and globus pallidus.…”

“…Following an exhaustive examination of the contents, 224 articles were excluded according to the selection criteria detailed in the method section. Of the 33 articles being selected that report iron content (summarized in Table 1), 11 of them employed postmortem analyses2491011121314151617, 14 were measured by R2* 318192021222324252627282930 and 8 by MRI relaxometry SWI3132333435363738. The disease comorbidity and diagnostic performance of the cohorts of these 33 studies are summarized in Table S1.…”

“…Devos, et al have recently reported very encouraging results showing that DFP improves PD motor performance and reduces nigral R2* values on GRE MRI, suggesting that DFP can be the first disease-modifying therapy for PD (127). Because QSM is superior to R2* for evaluating nigral iron (131,132,134,135,145), an important potential application of QSM in PD is to measure DFP's effectiveness of target-engagement in clinical trials.…”
