Reduction of the Endocochlear Potential by the New “Loop” Diuretic, Bumetanide

Abstract: The effect of bumetanide upon the endocochlear potential (EP) was examined in 46 guinea pigs. The EP was reduced with dosages of 5 mg/kg or more. The maximum depression of the EP (reduction to -30 to -40 mV) was obtained at a dosage of 30 mg/kg. The recovery of the potential was incomplete at any dosage within three hours and the response pattern of the EP to bumetanide was similar to that to ethacrynic acid. The present experiments revealed that bumetanide, by weight, has a stronger ototoxic potency than the … Show more

“…Slc12a2 is essential for the uptake of K + from cochlear and vestibular perilymph as well as for uptake of K + from the intrastrial space, which is essential for the generation of the endocochlear potential. Inhibition of Slc12a2 with loop‐diuretic abolishes the endocochlear potential (Kusakari et al 1978). K + secretion fails in mice that lack Slc12a2 , and endolymphatic spaces appear collapsed (Dixon et al 1999; Delpire et al 1999; Flagella et al 1999).…”

“…administration of succinyleholine chloride. The EP was recorded through the round window using the method described elsewhere (Kusakari et al 1978). Under an operation microscope, the larynx and the base of the skull were removed and an incision was made in the dura mater to expose the vertebral and basilar arteries.…”

“…They are thought to play pivotal roles in cochlear K ϩ circulation, because application of specific inhibitors for each apparatus reduced EP (24,43,47,48) and targeted ablation of the genes for NKCC1 and the K ϩ channels collapsed scala media (54,81,84). The K ϩ channel Kir5.1, expressed in the fibrocytes in the lower part of the ligament, may negatively regulate K ϩ circulation and may be important for precise control of K ϩ flow (23).…”
