Reduction of lesion in injured rat spinal cord and partial functional recovery of motility after bone marrow derived mesenchymal stem cell transplantation

Abstract: AIm:This study aimed to analyze the effect of rat bone marrow-mesenchymal stem cells (rBM-MSCs) delivery on lesion site after spinal cord injury, and to observe the functional recovery after transplantation. mAterIAl and methOds: MSCs were isolated from rat femurs and tibias. The experimental rat population was divided into four groups: only laminectomy (1); laminectomy+trauma (2); laminectomy+trauma+PBS (3); laminectomy+trauma+MSCs (4). Their motility were scored regularly. After 4-weeks, rats were sacrificed… Show more

“…The MSCs were transfected by Neon transfection System (Invitrogen/ Life Sciences) according to the method described previously [13]. The stable cell lines were maintained by continuous culture of transfected cells in the culture media with G418 (Gibco/Life Sciences; 200 μg/mL).…”

“…During the acute phase, neuronal necrosis, axonal disruption, blood supply insufficiency, edema and excitotoxic neurotransmitters are triggered. These changes continue into the subacute phase, during which ischemia, oxidative stress, lipid peroxidation and inflammation predominate (9,15,17,25). In this study, the effects of transplanting rAT-MSCs immediately (acute) and 9 days (subacute) p.i.…”

“…In order to determine the multipotency of isolated bone marrow mesenchymal stem cells, rBMSCs were harvested in the third passage and divided into three groups: adipogenic, osteogenic and neurogenic differentiation protocols were performed as described previously [26]. DMEM supplemented with 10% FBS, 1% Penicillin/Streptomycin were added to each group and incubated for 2 days for confluency.…”
