Reduction of infection risk mediated by co‐culturing Vibrio parahaemolyticus and Listeria monocytogenes in refrigerated cooked shrimp

Abstract: This study has revealed that interaction of V. parahaemolyticus and L. monocytogenes should be considered when quantifying risks posed by these pathogens during consumption of seafood products. © 2018 Society of Chemical Industry.

“…The overnight culture was then diluted with 0.1% peptone water (PW; Beijing Luqiao Technology Co., Ltd., Beijing, China) to an optical density (OD) value for 1.2 ± 0.02 (about 10 9 CFU/ml) at 600 nm (OD 600). The automated turbidimetric system Bioscreen C (Oy Growth Curves Ab Ltd., Raisio, Finland) was used to test the corresponding OD values at regular intervals (Niu et al, 2018b).…”

“…L. monocytogenes was streaked onto PALCAM (PALCAM; Beijing Land Bridge Technology Company, Ltd., Beijing, China) plates with selective additives and incubated at 37°C for 30 h. Individual colony was grown in 9 mL brain heart infusion (BHI, Beijing Land Bridge Technology Company, Ltd., Beijing, China) and incubated overnight at 37°C with shaking at 180 rpm. The bacterial concentrations were diluted to ∼8 Log CFU mL –1 for use (Niu et al, 2018).…”

“…L. monocytogenes infection may cause listeriosis (de Grandi et al, 2018; Radoshevich and Cossart, 2018) and the mortality rate is high where almost 1/3 infected cases will be fatal (Grace et al, 2011). V. parahaemolyticus and L. monocytogenes can coexist in different seafood products and in post-harvest processing sites and these are potential sources of cross contamination (Liao et al, 2015; Zhang et al, 2015a; Niu et al, 2018).…”

“…Afterward, the density of the bacterial suspensions was measured by BioTek Synergy2 (Winooski, VT, United States) at 600 nm (OD 600 nm ). When the OD 600nm values of V. parahaemolyticus are in the interval (1.2, 1.3), the bacteria concentrations are 9 Log 10 CFU/g ( Niu et al, 2018 ). Dead cells were obtained by heating suspensions at 85°C for 30 min.…”

“…Rapid detection technique can improve the management of microbial food safety and lots of detection methods were developed for V. parahaemolyticus ( Nordin et al, 2017 ; Wu et al, 2018 ; Yang et al, 2018 ). Standard detection for V. parahaemolyticus , which is time and labor intensive, consists of isolation, purification and identification based on thiosulfate-citrate-bile-salts-sucrose (TCBS) agar and Polymerase Chain Reaction (PCR) ( Niu et al, 2018 ). This method, however, fails to detect viable but non-culturable (VBNC) cells and V. parahaemolyticus has been shown to easily enter VNBC state as a response to adversity ( Coutard et al, 2007 ).…”
