Reduced chromatin acetylation of malignant salivary gland tumors correlates with enhanced proliferation

Abstract: This study provides the first insight regarding histone modifications in SGTs. Our results suggest that epigenetic mechanism, particularly hypoacetylation of histone H3 (lys9), might play a role in the behavior of salivary gland tumors. Also, our findings suggest that interfering with the acetylation pattern of tumor histones represents a potential novel therapeutic strategy for the treatment of SGTs.

“…Hyper-or hypomethylation of DNA may induce the development of tumors by regulating their components, such as proteins of the extracellular matrix, and it can cause chromosomal instability (Dawson & Kouzarides, 2012;Sharma et al, 2010). Epigenetic alterations, isolated or combined, may be involved in the development and progress of SGT (Aniello et al, 2006;Augello et al, 2006;Binmadi et al, 2018;Daa et al, 2008;Durr et al, 2010;Ge et al, 2011;Guimarães et al, 2016;Hu et al, 2011;Kaira et al, 2013;Ling et al, 2016;Nikolic et al, 2018;Schache et al, 2010;Shao et al, 2011Shao et al, , 2012Stephens et al, 2013;Tan et al, 2014;Wagner et al, 2017;Wang et al, 2015;Williams et al, 2006;Xia et al, 2018Xia et al, , 2013Zhang et al, 2007Zhang et al, , 2014.…”

“…This important epigenetic event has been associated with many human cancers, including tumors of salivary gland origin (Wagner et al, 2017). The enzymes responsible for this process are called histone deacetylases (HDACs), and when they are altered in normal conditions may lead to tumor development and progress (Aniello et al, 2006;Wagner et al, 2017;Wang et al, 2015).…”

“…Epigenetics represents a crucial player for tumor development. It has been observed that malignant SGC's chromatin were hypoacetylated compared to benign tumors . Moreover, acetyl‐H3 expression was inversely correlated with Ki67, providing insight that histone modification and epigenetic mechanisms may play a role in the behavior of SGC.…”
