Redox Regulation of Stem-like Cells Though the CD44v-xCT Axis in Colorectal Cancer: Mechanisms and Therapeutic Implications

Abstract: Colorectal cancer (CRC) is a common neoplastic disease and a frequent cause of death. Drug resistance is a major challenge to CRC treatment and stem-like side-population (SP) cells may play a key role in this resistance. Although it has been recognized that cancer stem cells may be affected by redox status, the underlying mechanisms for this effect and the roles of celllular redox adaptation and antioxidant capacity in CRC remain elusive. Our study shows that CRC SP cells are highly dependent on cellular GSH t… Show more

“…Transfection of the siRNAs was performed using Lipofectamine 2000 (Invitrogen, Carlsbad, USA) according to the manufacturer's instructions as described previously [21]. Briefly, growing cells seeded at 4 × 10 5 cell per well in a 6-well plate were transfected with 10 nmol siRNA.…”

“…4B). Since a CD44 variant has been shown to interact with and stabilize the cysteine transporter xCT [32, 33], we asked if xCT levels would correlate with CD44 following treatment with PP242. PP242 reduced both CD44 and xCT protein (Fig 4B).…”

“…Moreover, in a recent report, also PEITC selectivity to stem-like side population was associated with its capacity to modulate the redox status in these cells by increasing their cellular ROS levels, which reduced their growth in vitro and their tumorigenic ability in vivo [31]. Therefore, it is plausible that the ability of watercress extract to target colorectal CSCs and/or circulating tumor cells and to induce anoikis might be related with redox modification.…”
