Reconstituting regulation of the canonical Wnt pathway by engineering a minimal β-catenin destruction machine

Abstract: APC and Axin are key negative regulators of Wnt signaling in development and oncogenesis. They form a multiprotein complex targeting the key Wnt effector β-catenin for destruction. Essential components of APC and Axin required for their cooperative function are identified, and the data are used to design a minimal β-catenin–destruction machine.
