Recent progress on rapid SARS-CoV-2/COVID-19 detection by CRISPR-Cas13-based platforms

Abstract: The limitations of conventional diagnostic procedures, such as real-time PCR-based methods and serological tests, have led the scientific community to innovate alternative nucleic acid detection approaches for SARS-CoV-2 RNA, thereby addressing the dire need for increased testing. Such approaches aim to provide rapid, accurate, cost-effective, sensitive, and high-throughput detection of SARS-CoV-2 RNA, on multiple specimen types, and without specialized equipment and expertise. The CRISPR-Cas13 system function… Show more

“…Compared to the two-step assay, this single-step SHERLOCK assay could diagnose SARS-CoV-2 with reduced sample-to-answer time and equal sensitivity with optimized conditions [ 107 ]. Zhang et al combined RPA with SHERLOCK to detect the S gene and Orf1ab gene of SARS-COV-2 [ 108 ].…”

“…Recently, Tian et al described a droplet-digital Cas13a assay (ddCas13a) by harnessing the natural confinement effect, enhancing the local molecule concentration for highly efficient reaction or detection [44]. In order to potentiate the local concentration of target sequence and RNA reporter for the Cas13a system in cell-sized volumes, the authors employed a simple droplet microfluidic platform [44,45]. In this context, a mixture of target RNAs and Cas13a mixture is emulsified with oil into thousands of picoliter-sized droplet reactors.…”

“…In this context, a mixture of target RNAs and Cas13a mixture is emulsified with oil into thousands of picoliter-sized droplet reactors. The accumulated fluorescent signal from the collateral cleavage of a single RNA target-activated Cas13a is sufficient to illuminate a picoliter-sized droplet [44,45] (Figure 2). In these conditions, after recognition by a crRNA, a single-target RNA can induce cleavage of over 10 4 quenched fluorescent RNA reporters, thus resulting in a fluorescently positive droplet.…”

“…Application of rapid antigen testing, such as the current POCT of IgM/IgG and the ELISA technology, which targets viral antigens or antibodies, may provide fast and costeffective detection of HCoVs mostly at the early stage of infection. Zhang and colleagues recently developed a new diagnostic procedure called STOP (SHERLOCK Testing in One Pot) for SARS-CoV-2 rapid detection within 1 h [116]. Although the technology is still awaiting approval for clinical use, it is expected to remedy the high demand for a rapid test due to the rapidly increasing global spread of SARS-CoV-2.…”
