Recent developments in the hammerhead ribozyme field

Abstract: Developments in the hammerhead ribozyme field during the last two years are reviewed here. New results on the specificity of this ribozyme, the mechanism of its action and on the question of metal ion involvement in the cleavage reaction are discussed. To demonstrate the potential of ribozyme technology examples of the application of this ribozyme for the inhibition of gene expression in cell culture, in animals, as well as in plant models are presented. Particular emphasis is given to critical steps in the ap… Show more

“…The modified nucleotide is not among the conserved core of nucleotides. 8,9 It is known that any base pair can be accommodated at the modification positions. 9 Since 2AP forms Watson-Crick-type hydrogen bonds with uridine resulting in a base pair that is isostructural to A=U, a negligible structural perturbation is anticipated (Fig.…”

“…9 One method that has been used to protect the ribozymes from intracellular degradation is to connect them to a vector RNA such as tRNA. 20 Nevertheless, new approaches in using RNA as a vector to escort and chaperone ribozymes in vivo should be further investigated in order to improve this technique, which has shown great potential for improving human health and agricultural productivity.…”

“…In the present study, we designed and synthesized hammerhead ribozymes against caspase-12 mRNA under the control of a U6 snRNA promoter. The U6 SnRNA Pol III transcripts were normally retained in the nucleus and expressed at high levels, transcripts were protected at their 3'-ends and Rev-binding RNAs and had certain advantages (49). It is a strong promoter for the expression of small RNAs in vivo.…”
