Recent considerations in the use of recombinant interferon gamma for biological therapy of atopic dermatitis

Abstract: Introduction Atopic dermatitis (AD) is the most common inflammatory skin disease in the general population. There are different endophenotypes of AD that likely have a unique immune and molecular basis, such as those who are predisposed to eczema herpeticum, or Staphylococcus aureus infections. Areas Covered In this review, we highlight the endophenotypes of AD where reduced interferon gamma expression may be playing a role. Additionally, we review the potential role of recombinant interferon gamma therapy i… Show more

“…Treatment with recombinant interferon-γ is well-tolerated in patients with severe, unremitting atopic dermatitis, with clinical improvement reflecting decreases in absolute white blood cell and eosinophil counts [ 68 ]. This type of therapy shows particular promise among pediatric patients with atopic dermatitis, although further studies are needed to determine the effects of interferon-γ on the prevalence of skin infection [ 69 ].…”

“…ИФН-γ играет центральную роль в дифференцировке Th1-клеток [32]. ИФН-γ способствует созреванию/дифференцировке кератиноцитов, усиливает синтез церамидов, что содействует восстановлению кожного барьера [33,34]. Снижение продукции ИФН-γ связано с колонизацией кожи S.aureus [34].…”

“…67% of children ages 3 to 20 reported greater than 50% improvement, compared to 56% of adults ages 21–40 and 44% of adults ages 41–65 [101]. Side-effects included flu-like symptoms, transient transaminase elevation, and granulocyte suppression [102]. …”
