Recent Advances in miRNA Delivery Systems

Abstract: MicroRNAs (miRNAs) represent a family of short non-coding regulatory RNA molecules that are produced in a tissue and time-specific manner to orchestrate gene expression post-transcription. MiRNAs hybridize to target mRNA(s) to induce translation repression or mRNA degradation. Functional studies have demonstrated that miRNAs are engaged in virtually every physiological process and, consequently, miRNA dysregulations have been linked to multiple human pathologies. Thus, miRNA mimics and anti-miRNAs that restore… Show more

“…Though the delivery of miRNAs and their antagomirs to target sites remains a challenge due to low cellular uptake and nuclease degradation, various delivery systems are still being tested. In particular, polymer-based non-viral vehicles have demonstrated advantages such as versatile structural modifications and protection of unstable miRNAs [261]. Poly(lactic and co-glycolic acid), chitosan, polyethyleneamine, and polyamidoamine dendrimers may also be carriers for efficient miRNA or anti-miRNA delivery systems.…”

“…Comprehensive reviews on miRNA provided an exhaustive update on miRNA delivery strategies, including virus-based delivery, non-viral delivery (artificial lipid-based vesicles, polymer-based or chemical structures), and recently emerged extracellular vesiclebased delivery systems, developed to overcome obstacles associated with cell-specific targeting, intracellular release, miRNA stability, and carrier-associated toxicity [94,95]. Few studies are currently available regarding the formulation of AdoMet-loaded nanoparticles, which resulted in an environmentally sensitive vehicle suitable for controlling AdoMet delivery [96,97].…”

“…The therapeutic use of miRNAs mainly includes both inhibition of pro-inflammatory miRNAs and overexpression of anti-inflammatory miRNAs. Viral vectors, lipids, polymers, inorganic and extracellular vesicles are usually used to deliver therapeutic miRNAs (Dasgupta and Chatterjee, 2021). However, due to the wide regulation of miRNA (Lewis et al, 2003;Broughton and Pasquinelli, 2016), how to reduce the regulation unrelated to the therapeutic purpose is the main problem at present.…”
