Reassessment of antioxidant activity of arbutin: Multifaceted evaluation using five antioxidant assay systems

Abstract: Arbutin, a practically used skin-lightening agent, has been reported to possess a weak antioxidant activity compared to that of its precursor, hydroquinone. However, its antioxidant activity has not been systematically evaluated. Hence, this study reassessed its activity using five assay systems. Assays were first performed using model radicals, DPPH radical and ABTS(*+). Arbutin showed weak DPPH radical-scavenging activity compared to that of hydroquinone, but showed strong ABTS(*+)-scavenging activity. Its a… Show more

“…The AA-2βG and AA scavenging reactions were rapidly affected at pH 3.0. Such findings are consistent with the results of others demonstrating the capacity of AA-2βG to efficiently scavenge DPPH in vitro (Takebayashi et al, 2008(Takebayashi et al, , 2010. .…”

“…As a natural AA analogue, AA-2βG has high antioxidant activity and may play pharmacological roles in antioxidant, antiaging, and antitumorigenic activities (Takebayashi et al, 2006(Takebayashi et al, , 2008(Takebayashi et al, , 2010Zhang et al, 2011). However, the AA-2βG antioxidative mechanism remains poorly understood due to a lack of in vivo investigations.…”

“…Arbutin has diverse functions, including that it is an anti-oxidant and a depigmenting agent [7]. In erythrocytes, arbutin shows long-lasting radical scavenging properties and also protects a membrane lipid from oxidative stress in human skin fibroblasts [8][9][10]. In cultured human melanoma cells (HMV-II and A375), mouse melanoma cells (B16), and a three-dimensional cultured human skin model, arbutin at concentrations below 300 lg/ ml decreased both tyrosinase activity and melanin content to 40 % of that in the control, thus blocking melanogenesis [11][12][13][14].…”

“…Recently, we reassessed the antioxidant activity of arbutin using five in vitro assay systems, though arbutin has been reported to possess weak antioxidant activity as compared to its precursor, hydroquinone. 12) We found that arbutin exerted strong antioxidant activity, comparable or even superior to that of hydroquinone in the ABTS þ -scavenging assay, the ORAC assay, and two cell-based antioxidant assays.…”
