Reaginic Antibodies on Basophils in Grass Pollen Allergy, as Shown by Rosette-Formation

Abstract: Antibodies to grass pollen allergens have been demonstrated on basophils of individuals with hay fever by means of a rosette-forming reaction with allergen-coated erythrocytes. The antibodies on the basophil cell membrane were shown to be IgE by inhibition of rosette-formation with specific anti-Ε serum. Not all persons giving positive skin reactions to grass pollen extracts had basophils capable of rosette-formation.

“…However, there is no correlation between mast cell ro settes and the reaginic content of sera, esti mated by PCA reactions. This discrepancy between cell-bound and circulating IgE has been reported and well documented in the rat [Jarrett and Stewart, 1973;Yamamoto et al, 1974] as well as in man [Aas and Jo hansson, 1971;Berg et al, 1971;Wilson et al, 1972]. This fact is of interest, inasmuch as clinical allergy mainly depends on cellbound reagins.…”

“…These rosettes result from ac tive sensitization and detect cytophilic anti bodies. Such rosettes have already been de scribed concerning macrophages [Boyden, 1964], basophils from guinea pigs [Wilson and Coombs, 1971] and basophils from al lergic individuals [Wilson et al, 1972]. Renoux et al [1975] described rosette-like aspects, when antihuman IgE-coated Sephadex granules were incubated with human IgE-coated rat mast cells.…”

“…To our knowledge, the only report based on the mast cell agglutination was made by Wii.son cl al. [16,17], who used an tigen-coupled erythrocytes in order to obtain an immune cyto-adhercnce with specifically sensitized basophiles. The use of antibody-coated Se-phadcx granules is another procedure which is rarely reported.…”

“…Methods have been reported to detect a specific allergen by the response of blood basophils to the allergen (Wilson, Marchand & Coombs, 1971;Wilson et al, 1972;Hirsch & Zastrous, 1972;Soifer & Hirsch, 1975;King, Alaye & Augustin, 1976). We described previously an indirect method in which the morphological changes of blood basophils following the addition of anti-IgE were observed on smear preparations (Kimura et al, 1975b).…”
