Reactive Oxygen Species-Responsive Nanoparticles Based on PEGlated Prodrug for Targeted Treatment of Oral Tongue Squamous Cell Carcinoma by Combining Photodynamic Therapy and Chemotherapy

Abstract: In this study, a reactive oxygen species (ROS)-responsive nanoparticle system was designed for combining photodynamic therapy (PDT) and chemotherapy for oral tongue squamous cell carcinoma (OTSCC)-targeted treatment. A PEGlated prodrug (RPTD) of doxorubicin (DOX) via thioketal linkage and cRGD peptide modification was synthesized and then used to prepare nanoparticles for encapsulating photosensitizer hematoporphyrin (HP). Thus, the obtained HP-loaded RPTD (RPTD/HP) nanoparticles had a regular spherical shape … Show more

“…In OSCC cells, RPTD/HP nanoparticles were effectively internalized and showed effective effects on cell growth inhibition and apoptosis induction after laser irradiation. In OSCC tumor-bearing mice, RPTD/HP nanoparticles show excellent tumor-targeting ability and significantly inhibit tumor growth through a variety of mechanisms after local laser irradiation [270].…”

“…They can control the activation of PSs by adjusting the distance of PS and quencher [154]. Many stimuli, such as external stimuli (e.g., light [155]) and internal environment of tumor (e.g., pH [135,139,156,157,158,159,160,161,162,163], enzyme [91,164], GSH [165,166,167,168], and ROS [169,170,171,172,173,174,175,176]), have been used as the keys to turn on the PSs by increasing the distance among the PSs.…”

“…Similarly, Pu and co‐workers reported a thioketal‐linked nanoparticle with mPEG polymer with an efficient level of cellular uptake and good bioavailability. Another self‐assembled nanoparticle based on thioketal‐linkages, in which the photosensitizer hematoporphyrin (HP) was encapsulated, has been developed by Wang and co‐workers . Combined chemo‐ and photodynamic therapy upon laser irradiation was observed in vivo.…”
