Pentagamavunone-1 (PGV-1), a curcumin analog, is a promising anticancer candidate for several cancers that have been proven in vitro and in vivo. However, the efficacy of PGV-1 against breast cancer is subject to improvement to achieve a more suitable application. Here we propose hesperitin, a citrus flavonoid, to increase the anticancer potency of PGV-1 in luminal breast cancer cells. We use the T47D cell as the model to investigate the effect of co-administration of PGV-1 and hesperitin on cell cycle block, apoptosis modulation, and senescence phenomena. PGV-1 and hesperitin showed strong and weak cytotoxicity with an IC50 value of 2 µM and 100 µM, respectively. The co-treatment of PGV-1 and hesperitin resulted in strong synergistic effects with combination index (CI) value of ≤ 0.2. This combination caused apoptosis in correlation with cell cycle disruption in G2/M phase at 48 h. In particular, PGV-1 and hesperitin combination increased the incidence of cellular senescence significantly higher than the single treatment. Despite its senescence potentiation, hesperitin did not induce senescence in normal cells. Taken together, hesperitin may increase the anticancer potency of PGV-1 by modulating cell cycle arrest and apoptosis via the senescence mechanism.

BACKGROUND: Cardamom (Amomum cardamom) essential oil (CEO) contains monoterpenes with antioxidant activity and is reported to exhibit anticancer activity against some cancer cell lines. Triple-negative breast cancer (TNBC) has the lowest prognosis among breast cancer types due to its aggressive characteristics. This study was conducted to explore the potency of CEO in inhibiting 4T1 cell proliferation and migration and compared its activity to sappan (Caesalpinia sappan) wood extract (CSE).METHODS: We used the 4T1 cell line as the TNBC cell model and tested the cytotoxicity of CEO by using a trypan blue exclusion assay. We studied the senescence induction ability of CEO using SA-β-Gal assay, the migratory inhibition activity using scratch wound healing assay, and inhibition of matrix metalloproteinase 9 (MMP-9) expression using gelatin zymography. RESULTS: CEO showed cytotoxicity toward 4T1 cells with the IC50 values of 60 µg/mL. CEO at ½ IC50 and IC50 concentration significantly increased cell senescence, but CSE did not. CEO at IC50 also reduced cell ability to migrate and also considerably reduced MMP-9 activity. Moreover, these activities related to the inhibition of the cell metastasis process were weaker compared than that of CSE.CONCLUSION: CEO showed potency as a chemopreventive agent on the TNBC 4T1 cell line model with moderate cytotoxicity. CEO induced 4T1 cell senescence, inhibited cell migration and suppressed MMP-9 expression.KEYWORDS: Amomum cardamom, Caesalpinia sappan, 4T1, senescence, cell migration, triple-negative breast cancer

Rice is an essential crop, growing in nearly every part worldwide. Grains are the most common rice plant part consumed as food; however, other plant parts also have some significant values apart from grains. Rice bran is the outer layer of the rice grain, an excellent source of various phenolic, flavonoid, and phytoactive compounds. This review article discusses the various compounds found in rice bran with their structure and properties. Different medicinal properties of rice bran are also discussed in this review article. Several constituents of rice bran show anticancerous activity through various mechanisms like cell cycle arrest and regulation of apoptotic factors. Anti‐inflammatory, antidiabetic, antihypertensive, and cholesterol‐lowering are the other activities of rice bran, briefly explained in the article, along with their mechanism of action. A bibliographic network, including keyword bibliography analysis and a three‐field plot, has also been discussed to show the trends and network of common keywords in the recently published documents in the Scopus database. Overall, this review explores the rice bran compositions with their structures, activity, and various medicinal uses of rice bran, which can be proven an efficient oil source for nutritional purposes.
